Taxol is a kind of diterpenoid compounds which has unique anti-cancer mechanism and anti-cancer effect,which is isolated from the plant of Taxus.Because of having basic skeleton of taxol,6-8-6-4 cyclic taxane diterpenoid compounds have been paid more and more attention and can be transformed into paclitaxel through effective methods.At present,the common analysis method of paclitaxel substances in natural substances as following:ethanol extraction,the mixture solvent with water and dichloromethane shock layering,then evaporated dichloromethane layer,adding n-hexane and methanol shocking,taking methanol layer into the detector;Or the second method:after extraction and purification by solid-phase column,elution with a large number of solvents,concentrated into high-performance liquid separation detector for detection.These processes are labor intensive and environmentally unfriendly.In this paper,a multi-component separation system composed of carbon fiber and activated carbon fiber columns in parallel was used to establish a method for on-line screening of trace taxanes in various natural products,and seven targets,including 10-deacetylbaccatin ?,baccatin ?,7-xylose-10-deacetyl taxol,7-xylosyl-taxel,cephalomannine,taxol,and 7-epi-10-deacetyl taxol were achieved on-line rapid analysis by tandem mass spectrometry(MS/MS)at the same time.The natural samples were extracted with methanol as solvent by ultrasonic and filtered.The extracts were directly introduced into multidimensional fractionation system.While the lipids,pigments and other weakly polar interferences were removed,targets were directly through the MS/MS for qualitative and quantitative analysis.The results showed that the method can quickly and accurately characterize taxol and its derivatives in complex plant samples with good accuracy and repeatability.The recovery of seven targets in pine needles was 75%-114%and the RSD was 1.40?-8.06%(n=6).The linear regression equation was y = 1.593x+44.883,R2 = 0.9994,the LOD was 25 ng g-1,the LOQ was 75 ng g-1,and the MDL was 100 ng g-1.It is a fast,efficient and reproducible method for the separation and screening of taxanes from complex samples in just 5 minutes.With less solvent(40 ?L)and easy to operate,it is also a green approach.This method provides a scientific and technical support for high-throughput,automated screening of new plant resources of taxol.
