Sulfonamides?SAs?is a genericterm for a class of drugswith p-aminobenzenesulfonamide.Sulfonamides is widely used in animal breedingproduction industries for its good antibacterial effect and low price.However,abuse of Sulfonamidesresulted in the drugs residues in animal production,which may lead to allergic reactions,poisoning and drug resistance even teratogenic,carcinogenic and mutagenic in consumers.Therefore,from the perspective of food safety,the detection of sulfonamide residues in animal-derived food is always an issue that people highly concerned.With the development of sulfonamides residues detection methods,LC-MS become the most commonly used method today with many advantages such as: wide selectivity,better sensitivity and high efficiency.However,the use of chloroform,acetone and other organic solvents made the detection method more health harmful,costly and environmental damage.This paper was tried to improve the detection method above with the purpose of using less organic solvent,reducing detection cost and increasing detection efficiency.In this paper,the effect of 5 kinds of extraction solvent?such as 5% formic acid-acetonitrile,5% aceticacid-acetonitrile,acetonitrile,trichloromethane,ethyl acetate?,7 extraction time?0min,5min,10 min,15min,20 min,25min,30min?,5 concentration temperatures?30?,40?,50?,60?,70??and 4 chromatographic column(AQ5?m 2.1mmI.D.×150mm,Agilent ZORBAX Eclipse XDB-C18 2.1×50mm,C18-MG ??pH 2-10?3?m 2.0mm I.D.×150mm,Waters ACQUITY UPLC HSS T3 1.8?m 3.0×50mm)on the result of sulfonamides residues detection was investigated.This study established a detecting method for 16 sulfonamides residues in animal derived food by using High-Performance Liquid Chromatography-tandem Mass Spectrometry?HPLC-MS/MS?.The linear correlation coefficient?R2?of 16 sulfonamides in the range of 1-50 ug/L was 0.99650.9994,the recoveries were 85.82109.51% with a relative deviation of 2.3011.25%,a detection limitation of 0.0030.022ug/kg and quantification limitation of 0.21.1ug/kg.The main procedure was as follow: The samples were extracted with 5% acetic acid-acetonitrile for 20 min,and then was purified by Bond ElutQuechers EM?for salt extraction?,EMR-Lipidstp and EMR-polish tubes in a simple and non-toxic way;and followed the purification,the samples were concentrated with nitrogen at 50 ?;C18-MG ??pH 2-10?3?m 2.0mm I.D.×150mm column was used as chromatographic column.In conclusion,when compared with the detection result of national standard method?GB/T 20759-2006?,the relative deviation was kept between 4.28% and 9.76%.Took the advantages of high accuracy,low cost,high efficiency and less organic solvent using into consideration,the detection method established in this paper is suitable for the detection of sulfonamide residues in animal-derived food.
