Reconstituted Tobacco,which is nearly equal to or even better than the natural tobacco in character,is made of some tobacco wastes,such as cabo and tobacco powder,by re-engineering process.The harmful composition to human-body(tar,nicotine,sugar content,combustion value,etc)of reconstituted tobacco is more easily controlled artificially,some spices can be added to change the taste.Compared to the natural tobacco which can not be controlled easily,reconstituted tobacco is a good choice to control and improve the quality of cigarette.At present,the water consumption of reconstituted tobacco production enterprises is larger,which is rich in nutrients,some harmful ingredients of cigarette and microbes.It is difficult and highly cost for the treatment of the sewage.Five domestic tobacco companies which produced Reconstituted Tobacco were investigated and the fact was found that the life-cycle of white water is about 3 to 7 days,in the last days of the cycle,it became sour and smelly,more sticky material began to aggregate,paper fractured easily at the paper machine,the production efficiency was reduced,which all had a bad impact on the quality of the product.The nutrient was enriched with the cycle,which led to the growth of microbes and polluted the production lines.In order to reduce water consumption,clean production,stabilize quality and improve the environment,the research in the number,types and rules of changes of bacteria in white water needs to be done.Compared with different methods of the bacterial detection,the best way is chosen and optimized to analyze the total number of bacteria in white water.This new method was used to study the change of bacteria,isolate and identify the main bacteria in white water.The contents and results are as follows:The verification of bacterial detection methods: This study selected flat colony counting method(FCCM)of national standard in the food industry and acridine orange direct counting(AODC)of national standard in ocean survey specification to tests the white water bacteria in the process of corruption.But this method could only detect the living bacterium which couldn't represent the quality of white water.Therefore,a detection method must be found which could detect both living and decay bacteria.After extensive literature review,combining with the production requirements and testing costs,we had a comparison in nephelometry?gravimetry?Methylene blue staining direct counting and Acridine orange direct counting(AODC).The result revealed that fines and aerosols in the white water could easily influenced the accuracy of nephelometry?gravimetry and Methylene blue staining direct counting,so they were not suitable for white water detection.AODC was used to stain the materials in the white water(except the bacteria),and it was found that AODC wasn't affected by the interfering substance in the white water.Then the method was optimized which was Formaldehyde fixed for 2 minutes,15 minutes of acridine orange staining.We combined with five domestic reconstituted tobacco production enterprises to analyze its repetition rate,reproducibility and precision,which demonstrated that AODC met the requirements of analysis.It could be expanded and had the potential to become the national industry standard.In conclusion,AODC is suitable for white water bacteria detection.The research in the change of bacteria's number in the circulation white water of reconstituted tobacco: AODC was used to study the law of the change of bacteria's number in the white water which is produced before technological improvement in the Xuchang Chip company.There was a rapid growth of the total number of bacteria in the white water in the initial production stage,peak around 25 h,the total number of bacteria reached dynamic equilibrium after continuous production of 36 h.After doing parallel experiments in the Hubei Chip company and Shanghai Chip company,it was found that the trend of white water's change was consistent with the result of Xuchang Chip company.On the basis of analyzing the rule of the bacteria change,and combining with production explain the reasons for the law.In the later half of the year in 2015,the technology in Xuchang Chip company used new technologies,such as centrifugation,flocculation,etc,which made the bacteria peak reduce by 52.2%,peak flowing out extending from 25 h to 40 h,white water's serviceable cycle extending from 3~5 days to more than 10 days.Isolation and identification of the bacteria in the white water.After dyeing the white water,the bacteria in the water was given priority to the gram positive bacterium.Four strains of dominant species are isolated in the white water.Gram staining found that these four types of bacteria were all gram positive bacteria and their shapes were mainly rod.Though 16 s rDNA sequence analysis,these four types of bacteria were bacillus subtilis(51.2%),bacillus licheniformis(18.5%),bacillus cereus(0.7%),flavobacterium(29.6%).The identification of white water bacteria provided data support for the next steps in project,which were screening the way of bacteria introduced and specific antimicrobial treatment.
