The dry sensation of cigarettes always restricts the development of cigarette industry.In order to reduce the crumb produced in the manufacture of cigarettes,it is necessary to add humectants in them.However the traditional humectants have defects of itself,so it is urgent to find a new natural type.The Area along the coast of Zhejiang is rich for seaweed resources.The polysaccharide in seaweed contains copious hydroxyl groups which are apt to form hydrogen bond with water and have moisturizing effect.This study was aimed to select the kind of seaweed with fine smoking test effect and great moisturizing performance and to test its optimization of process for extraction,separation and purification,and moisturizing performance.The main research contents and results are as follows:1.Screen the selected 35 seaweedsalong the coast of Zhejiang and on the basis of the smoking test results of the extracts.The extracts are obtained by way of centrifugalization and filter after ultrasonic extraction through 35 seaweeds.The result of the total sugar content test of the 35 kinds of seaweed indicates that the total sugar content is between 1%and 20%,and the difference of the total sugar contents of different seaweed is remarkable.The crude extracts of the 35 kinds of seaweed are sprayed over the tobacco in a fixed proportion.Three seaweed samples are selected on the basis of the sensory evaluation of the cigarettes made from the tobacco.2.Fix a Sargassum horneri with fine smoking test effect and utilize the Optimized Ultrasonic Response Surface Analysis to extract the Polysaccharides of Sargassum Horneri.The result of its nutrient components shows:protein 18.11%,fat 1.43%,carbohydrate 55.42%,and ash 17.82%.With this guidance,the study on separation and purification in the later period is carried out.Then the process for extracting polysaccharides of sargassum horneris is optimized by Response Surface Method.The optimum extraction condition is defined on the basis of the extraction rate of polysaccharides:the water-powder ratio of 56.7 ml/g,the power extraction of 480W,the distilled time of 25min,the duty cycle is 1:1.Under this condition,the theoretical extracting value of ultrasonic assisted extraction of polysaccharides from sargassum horneris is 10.82%,which is accorded with the actual extract value.3.Use radial flow coupling ultrafiltration to separate and purify the polysaccharides of sargassum horneris.The radial flow chromatography is utilized to remove the protein in the extract of sargassum horneris.Based on the the retention rate of polysaccharides,the removal rate of protein and the migration time,the optimum processing parameter of purification is:the sample flow-rate of 5ml/min,the elution flow-rate of 40ml/min,the sample volume of 100ml and the sample concentration of 5mg/ml,the deproteinization rate of 74.35%and the polysaccharide recovery rate of 80.75%.Furthermore,ultrafiltration is used to separate the polysaccharides of sargassum horneris through the radial flow chromatography by molecular weight with Ultrafiltration Membranes of 10KD and of 100KD.It comes to a conclusion that polysaccharides of sargassum horneris are centralized in two groups including the one below 10KD and the one above 100KD,in which the remaining protein can be further removed.4.The crude extract of sargassum horneris and the three groups of separated and purified polysaccharides are sprayed over the tobacco.By taking the distilled water as reference,the variation of the moisture content in the different time points are mensurated by difference method to evaluate their moisturizing performance.According to the result,compared with water,the polysaccharides of sargassum horneris can improve the moisturizing effect,and the moisturizing effect of the seperated and purified polysaccharide group is more powerful than that of the crude extract.
