The Effect Of Koumiss On Immunoregulation In Wistar Rats

Koumiss is a probiotic beverage with rich nutrition.Studies had shown that lactic acid bacteria(LAB)isolated from koumiss plays a positive role in immune regulation,it can enhance the phagocytic ability of macrophages,promote immune cell proliferation and affect secretion of some cytokines,but there are no comprehensive reports about immunoregulatory effects of koumiss.So,this study was aimed to evaluate effects of koumiss on four-week-old Wistar Rats'immunoregulation by detecting spleen index,T,B lymphocytes transformation rates,concentration of serum IgG and IgA,percentage of CD3+,CD4+,CD8+,CD4+CD25+T-lymphocyte subsets,calculating the ratio of CD4+/CD8+ and CD4+CD25+/CD4+.24 Specific Pathogen Free(SPF)Wistar Rats were random grouped into 1 control and 2 experimental groups,each group had 8,control was oral administrated 4mL sterilized physiological saline with basis daily ration,groups were oral administrated 4mL different koumiss with basis daily ration.Wistar Rats were executed by a cervical vertebrae luxation after four weeks,took spleen under aseptic condition and drawn blood sample from the heart.Weighed spleen to calculate spleen index,results were that group 1 and 2 were significantly higher than control(P<0.05).Detected T,B lymphocytes transformation rates through a thiazolyl blue tetrazolium bromide based reaction,results were that T lymphocytes transformation rates of group 1 was slightly higher than control,no significant difference,group 2 was significantly higher than control(P<0.05).B lymphocytes transformation rates of group 1,2 were slightly higher than control,no significant difference.Detected percentage of CD3+,CD4+,CD8+,CD4+CD25+T-lymphocyte subsets by flow cytometry and calculated ratio of CD4+/CD8+,CD4+CD25+/CD4 +,results were that CD3+,CD4+,CD8+ T-lymphocyte of groups improved by different degrees compared with control.Ratio of CD4+/CD8+ was closed to the upper limit within the normal range,And percentage of CD4+CD2 5+T-lymphocyte and the ratio of CD4+CD25+/CD4+ were significantly lower than control(P<0.05).Detected concentration of serum IgG and IgA based on enzyme linked immunosorbent assay,results were that serum IgG concentration of that group 1 and 2 were significantly higher than control(P<0.05).Serum IgA concentration of group 1 was slightly higher than control,no significant difference,group 2 was significantly higher than control(P<0.05).The study results showed that koumiss could up-regulate spleen index,T,B lymphocytes transformation rates,concentration of serum IgG and IgA,percentage of CD3 +,CD4+,CD8+ T-lymphocyte subset,and the ratio of CD4+/CD8+ is within the normal range,down-regulate percentage of CD4+CD25+T-lymphocyte and ratio of CD4+CD25+/CD4+ in Wistar Rats,may promote the proliferation and differentiation of lymphoid cells,enhance the body's humoral immunity and cellular immunity,thereby assisted immunoregulation of Wistar Rats.
