Purpose: Fermentation process and preliminary study on Pharmacodynamics ofGloeostereum incarnatum S. Ito et Imai in order to laying the foundation for thedevelopment of new drugs.Methods: Liquid fermentation of bio-engineering methods to study the fermentationprocess; carrageenan-induced rat paw swelling test to determine anti-inflammatory effectsof fermentation activity substances---crude polysaccharides; acetic acid-induced rat colitismodel test to interpret the role of anti-colon ulcer; test of vitro cultured lymphocyteenhanced to determine its immune enhancement.Results: Through the study on fermentation process, filter out the the optimal carbonsource is glucose in fermentation of Gloeostereum incarnatum S. Ito et Imai; optimal nitrogensource is Soybean powder and peptone; optimal Inorganic salts is KH2PO4and MgSO4.Its shaking flask seed-stage is5d;and putting the cans-stage is7d; Culture temperature is25±1℃a nd PH is6-7. Anti-inflammatory test, high dose group and medium dose group ofGloeostereum incarnatum S. Ito et Ima crude polysaccharide compared with the controlgroup, p<0.01;anti-colon ulcers and improving immunity test, high dose group comparedwith the control group, p<0.01,and medium dose group compared with the control group,p<0.05.Conclusion: Fermentation production technology parameters of Gloeostereumincarnatum S. Ito et Imai is controllable,and its crude polysaccharides of fermentationbroth has significant anti-inflammatory, anti-ulcer and enhancing the role of immunity,which can be used for anti-ulcer and enhancing the role of immunity drug screening. |
