Rutin is widely existed in fruits and vegetables,and it has been widely used in Chinese traditional medicine,because of antiviral,anti-inflammatory,anticancer,antibacterial,dilation of blood vessels and antigen biological activity.Rutin was a natural antioxidant with good prospect in food and medicine industry.But the application in hydrophilic food was limited by its poor water solubility.In order to solve this problem,this study used carboxymethyl-?-cyclodextrin(CM-?-CD)as material to embedded the rutin,then formation of inclusion with the moles ratio of 1:1 rutin-CM-?-CD.Response surface method was utilized to study mass ratio,embedding temperature and embedding time.The formation of rutin-CM-?-CD inclusion complex was identified by Sanning Electronic Microscopy(SEM)?FI-IR.And the antioxidant capacity was compared with unembedded rutin.The main results are as follows:(1)The response surface method indicated that the optium ratio of rutin and CM-?-CD was 1:4,the embedding temperature was 60?,and the embedding time was 2h.And Rutin is contained in the inner cavity of CM-?-CD in the form of a molecule,which is proved that the formation of the rutin-CM-?-CD inclusion is not new compound.(2)This paper investigated the solubility of rutin complex,which demonstrated that the rutin inclusion complex can improve the solubility of rutin reached 1.76mg/mL,and is about35 times than the buried solubility of rutin.In this study,the embedding method and embedding material can obvious improve the solubility of rutin.(3)This paper studied the antioxidant capacity of rutin-CM-?-CD.The differences of the ability of clearing DPPH free radicals for rutin,rutin-CM-?-CD and Vc in different concentrations of 1ug/mL,3ug/mL and 5ug/mL was studied.Results showed that:The highest clearing rates of DPPH free radicals of rutin,inclusion complex and Vc were 39.66%,46.29%and 58.46%under 1ug/mL concentration.The highest clearing rates of DPPH free radicals were 55.65%,67.48%and 71.32%under 3ug/mL concentration.The highest clearing rates of DPPH free radicals were 75.21%,84.35%and 86.67%under 5ug/mL concentration.Which indicated that the ability of clearing DPPH free radicals of rutin inclusion complex was significantly higher than rutin,and there was no significant difference compared with Vc.Research results of antioxidant effect of oil showed that:At the initial stage of antioxidation,the POV value of inclusion compound was 2.81mmol/kg,higher than V_E(2.02mmol/kg)and rutin(2.19mmol/kg),which indicated that the antioxidant effect was no significant in the stage.But from the third day,the POV value of inclusion compound was 3.27mmol/kg,slightly higher than V_E(3.12mmol/kg),lower than rutin(4.08mmol/kg).After thirteen days of detection,the POV value of the inclusion compound was higher than V_E and lower than rutin.Therefore,the antioxidant effect of the embedded rutin was higher than rutin.
