The name of the Lycium barbarum in the "Book of Songs",2000 years ago,has a long history of planting,in the field of TCM health care occupies a large place,is a homologous variety of edible medicine.It not only has high medicinal value,but also has good economic benefit.In our country,the planting area has been enlarged every year,and cultivated in Ningxia,Qinghai,Xinjiang,Inner Mongolia,Gansu and Hebei.However,due to the different ecological environment of various habitats,the content and safety have some differences.In the international trade market,the export of Chinese herbal medicines accounted for only 1% of China's total exports,because its quality is not up to the European and American countries quality standards.The quality standard of Medlar in the 2015 edition "Chinese Pharmacopoeia" is character identification,water,ash,impurities and other routine items of inspection,safety Check for heavy metals(lead,arsenic,cadmium,mercury,copper),content determination only Lycium barbarum polysaccharide and betaine two components,and the determination of betaine is thin layer scanning method,there are many external interference factors,Because of the problem of poor separation effect and low reproducibility,based on the above situation,this research has been improved on the basis of the existing standard of Chinese pharmacopoeia to improve and improve the quality control level of Medlar.1.To increase the content determination of rutin in flavonoids by high performance liquid chromatography,after the selection of extraction method and optimization of chromatographic conditions,70% ethanol heating reflux extraction was determined,the flow phase was methanol: 0.5% phosphoric acid solution gradient elution,ultraviolet detector detection,detection wavelength 358 nm,Kromasil ODS-3 chromatographic column.Results the theoretical plate of flavonoids was more than 3000,the separation degree between main component peak and adjacent peak was greater than 1.5,the stability,precision and repeatability were good,and the result was accurate and reliable.2.To increase the content of carotenoids and determine the content of ?-carotene,zeaxanthin and zeaxanthin by HPLC method by searching the literature optimization method.Experimental results The number of ?-carotene was more than 3000,and the separation between 13 peaks of carotenoids was more than 1.5,and the results of methodological study showed that the precision,repeatability,accuracy and stability were good.3.Increased organochlorine pesticide residues detection,the use of gas chromatography to detect 666,DDT,five chloro-nitrobenzene,Ai reagent,di reagent,the content of different agents.The experimental operation is simple and the pesticide residues can be separated effectively.4.The detection method of Betaine was modified to a more advanced high-performance liquid chromatography method.To find the literature,compared the ultraviolet detector,the evaporation light scattering detector,the amino chromatographic column and other chromatographic conditions,the results show that: the existing evapotranspiration detector for the low-temperature detector,through the transformation of the detector parameters,still can not meet the needs of the quantitative detection of betaine.Using amino column and C18 chromatographic column,the ultraviolet method can not effectively separate the detected components from the impurity peaks.Finally,the hydrophilic hilic chromatographic column was used to separate the samples and the ultraviolet detector was tested and the separation effect was better.5.The determination of heavy metal content of Lycium barbarum from different habitats,and the detection method of arsenic and mercury by atomic absorption spectrophotometry is improved by atomic fluorescence spectrometry,the experiment is simple and feasible,and the result is accurate and reliable.The study of quality control of Lycium barbarum L.Can be used to increase the market competitiveness of medlar products and promote the export trade of Chinese Medicine.
