Nesfatin-1 is a kind of polypeptide which is functional diversity,it involved in many physiological activity and plays a certain role in various systems of animal body.Currently,it,however,is just restricted to rodents,goldfish,there's no research report for canine species.Research object in this experiment was baby,childhood,and puberty female dogs and adult female and male dogs.Immunohistochemistry technique was used to detect the distribution of Nesfatin-1 in the reproductive axis of adult dogs,and RT-PCR was used to detect the quantity of NUCB2 m RNA in the reproductive axis of female dogs during different growing stages.Aim is to initially elaborate the expression pattern of Nesfatin-1on the reproductive axis of the dog.Through comparative analysis determine the exact location of the distribution of Nesfatin-1,different development phase content change rule,to determine Nesfatin-1 the influence of distinct developmental stage,provide reliable theory basis for clinical treatment of diseases.Normal adult male and female dogs all the three buying from the market.Raised after a week did not locate significant infectious disease,blooding for dead.Treatment in the hypothalamus pituitary and tests in 4% paraformaldehyde fixed place 24~48 h after embedding,the conventional paraffin slice,thickness of 5 microns,and then to hematoxylin immunohistochemical staining microscope staining microscope and photographed.The test results show that the immune Nesfatin-1 positive cell in adult reproductive axis has different degree of distribution.In the hypothalamus,SON,LHA area have positive immune cells,the pituitary gland of neurohypophysis and adenohypophysis was positive,ovary oocytes and testicles in contorted senimiferous tubules.Fluorescence quantitative PCR results showed that in different development stages of ovary and pituitary NUCB2 mRNA in infancy,along with the development of early to early love,in the early period NUCB2 mRNA levels rising,reached the highest in the early period of the content after entering adulthood NUCB2 mRNA content is gradually decline,different development stages in the same organization NUCB2 mRNA levels were significantly different(P < 0.01).And NUCB2 mRNA content in the hypothalamus from infancy to adulthood in the rising trend,and the different development stages of NUCB2 mRNA content in the same organization is also extremely significant difference(P < 0.01).Three reproductive axis NUCB2 mRNA content in the ovaries is the highest in four stages.According to the result of the above can be concluded that the Nesfatin-1 exists not only the central nervous system and the peripheral system.Because the NUCB2 mRNA in the early period of content have a significantly higher degree of speculation from previous Nesfatin-1 May regulate the mood at the early start of dogs.Ovary NUCB2 mRNA content in the highest,prompt the ovaries to produce Nesfatin-May 1 or its storage organs.
