In recent years, Genetically Modified crops had large-scale cultivation in worldwide. With the increasing scope and cultivation area, its potential ecological security problems brought more and more concerns and discussion. Soil is an important place for crop plants and environment,they can influence each other.So to illustrate whether it would affect the environment, we must first explore whether it would bring obvious change to environment of rhizosphere soil.In this study, experimental materials is TaDREB4 transgenic wheat TB4.Detecting the soil enzyme activity, fungi, bacteria quantity, the diversity of microbial communities of its rhizosphere soil.TB4 is developed by the Institute of Crop Sciences, Chinese Academy of Agricultural Sciences, showed better drought tolerance in the previous study. TB4 and its receptor Jimai 19 sowed in the experimental base in Jinan, Shandong Academy of Sciences, in October 2012.At the same time, in order to more effectively explain wether it has significantly affected, the sowing condition divided into natural conditions and drought stress. Sampled in its sowing, reviving, filling, mature growth stage, taking five-point sampling method, TB4 and its receptor Jimai 19 both have four repeated.The main results are as follows:1. Measuring the urease, dehydrogenase and invertase activity in rhizosphere soil to detecting the change of soil enzyme activities, respectively, using phenol hypochlorite colorimetric method, TTC-dehydrogenase test, DNS colorimetric method.The experimental results showed that: under natural conditions and drought stress conditions,urease activity has no significantly different in the four growth stages, so does the dehydrogenase activity and urease activity.2. Measuring he the number of Pseudomonas fluorescens and Fusarium which is common in soil. Quantitative PCR method is used to quantitative analysis.Pseudomonas fluorescens and Fusarium quantitative analysis showed that the TaDREB4 transgenic wheat TB4 and its receptor Jimai 19 in natural conditions and drought stress conditions have no significant difference in the four growth stages, the quality of Pseudomonas fluorescens between the period showed a gradual upward trend, which may be associated with a change in temperature;the quality of Fusarium between the period showed that first increased and then decreased, which is in line with the development of the disease process.3.Using PCR-DGGE to analysis diversity of microbial community.The experimental results showed that the diversity of microbial community which in rhizosphere soil of TaDREB4 transgenic wheat TB4 and its receptor Jimai 19 have no significant difference, so as its diversity index and evenness index.With all these showed that TaDREB4 transgenic wheat TB4 has no significant effect to its rhizosphere soil,its cultivation is safe and reliable.The study was carried out not only providing basic data for the evaluation of the environmental safety of gene transfer wheat, but also lay the technical foundation of safety assessment of genetically modified.
