Carassius is an important reared freshwater species, which has great market prospects for the prolific nutrition facts. Allogyogenetics crucian carp and Allogyogenetics crucian carp (no.3) are main varieties in Aquaculture. To compare growth performance and nutritional value of Allogyogenetics crucian carp and Allogyogenetics crucian carp (no.3), growth comparison experiment was Implemented. To study the Correlation between morphometric attributes and body weight, morphometric measurement was taken from 152 individuals of Allogyogenetics crucian carp (no.3) and 121 individuals of Allogyogenetics crucian carp. Single nucleotide polymorphisms(SNPs)in MSTN gene were identified and genotyped by SNaPshot analysis. SNPs which associated with growth traits were also analyzed. Main research results as follows:1. The attributes used in growth performance compare include body weight, body length, body width, body depth, AGRw, AGRBL,AGRBW,AGRBH, percentage of unfilled grainsand fatness. The results showed that Allogyogenetics crucian carp has faster growth rate than Allogyogenetics crucian carp (no.3). Correlation analysis between morphometric attributes and body weight shows that body length has the maximum correlation coefficient with body weight. The nutritional value compear between two varieties shows that Allogyogenetics crucian carp has advantage in total amino acids, essential amino acid content and delicious amino acid, and Allogyogenetics crucian carp (no.3) has advantage in fatty acid content.2. In order to clone full-length MSTN gene, homology cloning and rapid-amplification of cDNA ends(RACE) were used to acquire full-length DNA. A full-length MSTN DNA sequence in Allogyogenetics crucian carp (no.3) and prat of MSTN DNA sequence in Allogyogenetics crucian carp was cloned. The similarity betweenthem was 99.1%. Semi-Quantitative RT-PCR results indicated that MSTN gene expressed in the 6 tissues-brain, muscle, muscle, gills, heart, intestines and liver. The MSTN transcript was detected at a high level in brain, moderate in muscle, gills, and low in hesrt, intsetines and liver.MSTN had high expression in brain of Allogyogenetics crucian carp, indicating that MSTN gene in Allogyogenetics crucian carp might not only play roles in muscle growth and development but also has important role in neural development.3 SNPs in exon2 and exon3 of MSTN gene were find by DNA sequencing.3. Take 152 individuals of Allogyogenetics crucian carp (no.3) and 121 individuals of Allogyogenetics crucian carp as research object, genotypes of three sites were selected from MSTN gene using the technology-SNaPshot. There are two mutational sites in exon2 and exon3 respectively of MSTN gene of Allogyogenetics crucian carp (no.3) and one mutational sites in exon2 of MSTN gene of Allogyogenetics crucian carp. Correlation analysis between polymorphism of the mutational site and growth traits was carried. The results showed that the site of different genotypes in Allogyogenetics crucian carp (no.3) has significant differences on weight gain,body length. The body length of GT genotype is significantly higher than TT genotype (P?0.01). The weight gain of GT genotype is significant higher than TT genotype (P<0.05).But the site of different genotypes of Allogyogenetics crucian carp has no significant differences on growth traits. GT individuals had an advantage which could be a molecular marker for growth traits selective breeding in Allogyogenetics crucian carp (no.3). But further experimentation is necessary to verify the accuracy.
