Huanjiang miniature pig is a famous local variety in Guangxi, which is Chinese rare breeds, and through the national geographical indications of origin certification in 2003. Farmer and consumers are interested in excellent properties such as resistance to crude feed, cold, prolific, fresh meat and motherhood. However, Huanjiang pig and other small pig breeds are ignored since the reform and opening up, China has attached importance to solving the problem of food and clothing, aquaculture tends to feed large and grow fast pig species. In addition to the Huanjiang pig diarrhea piglet mortality, resulting in less amount of Huanjiang pig breeding and high cost. At present, the Huanjiang pig related functional gene research is rarely reported, the study bassed on candidate genes related to the ability of anti diarrhea and the litter traits of sows, including MUC13, FSHp, ESR, and ZAR1. The polymorphism detection of these candidate genes by PCR-RFLP, HRM and direct sequencing assays. The details of results were described as follows:The polymorphism detection of MUC13 related to piglet diarrhea resistance/susceptibility trait in Huanjiang pig. The results showed that four of the five analyed SNPs A, B, C, and D loci have polymorphism in Huanjiang pig, including SNP A32847G, SNP A37731G, SNP A42824C and SNP A46592G respectively. There was no polymorphism detected in E site. According the reports that type GG and CC are resistance genotype, the genotype frequency of 154 pigs, which AA and AG genotype frequencies of A locus detected were 0.792 and 0.208, favorable G allele frequency was 0.104; CT and TT genotype frequencies of B locus detected were 0.136 and 0.864, favorable C allele frequency was 0.068; AA?AG and GG genotype frequencies of C locus detected were 0.694?0.279 and 0.027, favorable G allele frequency was 0.166; GC and CC genotype frequencies of D locus detected were 0.7344 and 0.656, favorable G allele frequency was 0.172. The favorable allele gene resistance frequencies are low in Huanjiang pig, which was consistent with the investigation of diarrhea mortality in Huanjiang neonatal and young pigs. And there were significant differences in the distribution of different genotypes of four loci in population health groups, piglet diarrhea piglets and Piglet diarrhea death groups(P<0.05), what's more, the C locus had extremely significant difference(P<0.01), which shows that different genotypes are associated with resistance or susceptibility of piglet diarrhea.Polymorphism analysis on FSH? ESR and ZAR1 in Huanjiang pig. The results showed that the first successful cloning and sequencing of Huanjiang pig ZAR1 gene exon 3 and ZAR1 gene in GeneBank (DQ231443, gi:83727928) of exon 3 share the same sequence each other. The polymorphism gene mutations of FSH? and ESR were not found in 134 test samples. Five SNP sites were found by DNA sequencing in ZAR1 gene Exon3 and part of intron3, but there were no C54T mutation in exon3 which was reported that have significant impact on litter size. The result shows that the different varieties of pig have different patterns in gene mutation polymorphism, and this finding has important implications for the establishment of genetic marker-assisted selection in Huanjiang pigs.
