Phenacoccu ssolenopsis Tinsley is a new kind of plant invasiveand quarantine pests.They are widely distributed in 15 provinces across China.It is difficult to control effectively by chemical pestcides because of its wide host plants range and strong fertilityand complex waysin spreading.P.solenopsis,covered withsilty in its back,which has caused seriously harm in cotton,vegetables,and ornamental plants and so on.In order to explore the population dynamics and adaptability in different hosts of P.solenopsis,this study sets four temperature level(30?,26?,22?and 18?)and three kinds of hosts(cotton,tomato,and hibiscus)to examing its influences on growth and reproduction of P.solenopsis,furthermore,we established its transcriptome.Our study will provide the scientific basis for the prevention and control for P.solenopsis.These experimental results were summarized as follows,1.In the selected three kinds of host,P.solenopsis development starting point temperature on tomato was higher than on other two kinds of host.However,its effective accumulated temperature is lower relatively.The population trend index of P.solenopsis,which fed on cotton and tomato at 30?,on cotton and tomato at 26? and on tomato at22?,were significantly higher than other processing.Its value depends on appropriate host,high spawning,suitable temperature and low mortality rate.Net proliferation rate(R0)increases with the rise of temperature.It showed that its biggest value at 30?.But it has difference between different kinds of host.The biggest R0 valueat 30? is 72.36 which fed on cotton,while the biggest R0 value at 26? is 74.36 which fed on tomato.The R0 value of P.solenopsis fed on hibiscus was lowest under four temperature level.The value of intrinsic increase rate(rm)and finite increase rate(?)is higher of P.solenopsis fed on cotton and tomato at 30?.The doubling time and the average generation rate value showed that it extended with the temperature decrease.This study found that temperature were the main factors to impact P.solenopsis growth and development.At the same time,host for the influence of the temperature effect had the effects of a certain degree of dilution.The 26?~30? condition is more suitable for P.solenopsis fed on cotton and tomato.2.In the selected six kinds of host conversion under four temperature level,the results showed that,at 30?,the longest generation times of female adult,male adult and all of them appear in the experimental group that P.solenopsis fed on hibiscus swithingto cotton or tomato swithing to cotton.This phenomena further prove that temperature on the influence host of the effect has the effects of a certain degree of dilution.By hosts conversion,its development starting point temperature present the tendency of decrease compared with single host.The development starting point temperature value of pupa falling from 19.48? to 7.18?,3nd instar nymph value decline from 17.70? to 6.00?.On the contrary,the effective accumulated temperature value has a tendency to decline.At the same temperature,the value of net proliferation rate(R0)has stronger influence among different hosts conversion compared with fed on one host.The value of intrinsic increase rate(rm)and R0 have some positive correlation.At 30?,the biggest rm value is0.19 appeared on tomato swith to hibiscus.But its value relatively small when it fed on hibiscus swith to tomato and hibiscus swith to cotton,Its value respectively 0.11 and 0.09.This study found that,individual development duration compared with transformation before,its value is extened.However R0 value declined.3.This study analyzed and established the transcriptome sequencing of female adult and nymph of P.solenopsisusing the high throughput sequencing platform,and assembled the sequencing reads into Unigene using the de novo strategy,and conducted annotation of GO functional classification and KEGG pathway analysis to the unigene.The abundance and significant differentially expression of genes in P.solenopsis were calculated using RPKM way.We obtained 48223398 pairs raw data reads of nymph(about 7.23 G)and 85298908 pairs data reads of female adult(about 12.79 G).There were 57456 transcripts and 43198 genes reached 45459125 bp.The longest was 21289 bp,the shortest was 201 bp.The N50 value was 1776 bp,which showed patchwork genes quality was better.The 43198 Unigenewere annotated using the blast tools,and 14416 of which were annotated to Swiss-Prot,22397 to NR,16874 toPfam,11572 to KEGG,15581 to KOG and 13190 to GO.The functions of 13190 to GO were transcription,transcriptional regulation,translation,cell division,autophagy,and signal transduction and so on.The functions of 11572 to KEGG were organismal systems,metabolism,genetic information processing,environmental information processing,and cellular processes.There were 71 genes associated with its environmental adaptation.Compared with nymph and female adult,there were 2303 significant differently expression genes.Among them,the expressiond of 93 differently expression genes were increased,and2210 were reduced.Ourtranscriptome study between nymph and female adult of P.solenopsis obtained a lot of genetic data,and can provide a solid foundation from two levels of genes and protein research for the external environment conditions.
