Rehmannia glutinosa f.Hueichingensis(Chan et Sehih)Hsiao is a perennial herb belonging to the family Scrophulariaceae,one of the Chinese famous “Four Huaiqing Chinese medicines”,widely distributed in Wenxian,Wuzhi and Qinyang of Henan,etc.But the phenomenon of mixed varieties,virus infraction,low yield and quality,the high cost of the direct application of virus-free test tube seedling has become an urgent problem to be solved in the production of Rehmannia glutinosa f.Hueichingensis(Chan et Sehih)Hsiao.Based on the previous studies,this paper makes a further optimization of three levels of breeding phase system of Rehmannia glutinosa f.Hueichingensis(Chan et Sehih)Hsiao(producting of foundation seed,original species,production seed),and has carried on the cost accounting,virus detection was carried out on the different generations of virus-free seedlings grown in the field,the yield and quality of different Rehmannia glutinosa f.Hueichingensis(Chan et Sehih)Hsiao were compared,the prupose is to lay the technical and theoretical foundation for the factory production and application technology of Rehmannia glutinosa f.Hueichingensis(Chan et Sehih)Hsiao virus-free seedling.The results were as follows:1.Optimization of the technology system of three levels breeding phase of Huai Rehmannia glutinosa virus-free seedlingIn the production of foundation seed,optimize the conditions of rapid propagation,rooting and transplanting.The suitable basic medium for rapid propagation is MS + NAA 0.02 mg/L + 6-BA 0.20 mg/L + cold carrageenan 6.00 g/L + white granulated sugar 30.00 g/L,the best inoculation density is per bottle inoculated with 5 plants,and the best inoculation site is terminal bud;The suitable temperature for the rooting of different varieties plantlets of Huai Rehmannia glutinosa is 27?;The suitable medium for varieties plantlet transplanting is nutritional soil and vermiculite ratio is 1:1.In the production of original species and production seed,the best planting density is 20000 plants/mu,the best planting parrerns are 11 cm spacing,30 cm row spacing.2.The cost accounting at each stages of the technology system of three levels breeding phaseThe cost accounting of virus-free seedling obtained from the technology system of three levels breeding phase was carried on,contents include depreciation of fixed assets,components of culture medium,consumables,land rent,labor cost and water and electricity cost,etc.In this experiment,the cost of the foundation seed is 94.37 yuan/100 plant,the cost of the original species is 6.26 yuan/100 plant,the cost of the production seed is 1.93 yuan/100 plant.In the production of the foundation seed,the depreciation expense of fixed assets is the hightest cost,reaching 42.91%.In the production of the original species and the production seed,labor cost cost is the highest cost,61.24% and 56.45%,respectively.3.The comparison of the field yield,quality and the situation of the virus reinfected of the virus-free seedlings of different generations.For the first time,Virus detection was carried out on the field grown leaves of the virus-free one,two,three,four generations,the frist generation and the second generation were not infected with the virus,from the third generation began to infect the virus,the infection rate of the third generation was 60%,and the infection rate of the fourth generation was 100%.With the rate of virus infection increase,the field yield,the content of catalpol and verbascoside of the virus-free one,two,three,four generations were gradually decreased,among them,the field yield was 5603.90 kg,4359.50 kg,3992.50 kg,3258.20 kg,respectively.4.The comparison of field yield and quality of different varieties of Huai Rehmannia glutinosaThe difference of field yield between plantlets of different varieties of Huai Rehmannia glutinosa is relatively large,the order of field yield from high to low is "85-5","9302","Number 3 of Beijing","Jinjiu","Disease-resistant","Qinhuai",amoug them,the field yield of "85-5" is the highest,reaching 4837.00kg;The effective components of different varieties are different,the content of catalpol of "Qinhuai" is the highest,reaching 3.95%,the content of verbascoside of "Number 3 of Beijing" is the highest,reaching 0.0957%.5.The comparison of the planting density of the virus-free seedlings of "85-5" of Rehmannia glutinosa f.Hueichingensis(Chan et Sehih)HsiaoA comparative study of the field yield of the five field planting density of the virus-free seedlings of "85-5" of Rehmannia glutinosa f.Hueichingensis(Chan et Sehih)Hsiao is carried out,the results show that the field yield is the highest when the planting density is 20000 plants/mu,reaching 6105.80 kg,higher or lower than this density,the field yield will decline.
