In this study, the effects of the suitable amount of Aerosol OT and its effects on the daily amount of feed intake, nutrient digestion and metabolism of Small Tail Han sheep was studied by the combination of artificial rumen gas production and digestion and metabolism test. In experiment 1, under the conditions of two kinds of matrix of corn straw and mixed concentrate, to study the effects of 0, 0.05, 0.10, 0.15, 0.20, 0.25, 0.30, 0.35, 0.40, 0.45 mg/ml Aerosol OT(AOT) concentration on artificial rumen gas production; In experiment 2, Five of 3 year's old Small-tail Han non-pregnant ewes, with body weight of 50.3±1.7 kg, were fed with same diet(concentrate:roughage=4:6) and administered with aerosol OT(AOT) 0, 0.8, 1.2, 1.6 and 2.0 g/kg of diet respectively by Latin square design of 5×5 for the digestion and metabolism experiment; to study the effects of the administered doses of AOT on the voluntary intake, nutrient digestion and metabolism of sheep for determining the optimal dosage of AOT. The results showed that In experiment 1: the concentration of AOT affected the production of rumen gas, and the gas production with AOT concentration showed a parabolic change, At the AOT 0.20 mg/ml concentration, both the corn straw substrate and the fine feed medium were the largest, 12% and 14.4% higher than the control respectively(P <0.05). In experiment 2: Feeding 0.8, 1.2, 1.6 and 2.0 g AOT/Kg of diet, The feed intake of dry matter in Small Tail Han sheep was increased by 6.0%(P<0.05), 18.3%(P<0.05), 9.4% and 5.7% respectively; Apparent digestibility of dry matter increased by 4.4%(P<0.05), 5.0%(P<0.05), 2.1% and 0.5% respectively; The apparent digestibility of dry matter increased by 4.1%(P<0.05), 5.0(P<0.05), 2.5% and 0.1% respectively; Cellulose digestibility was increased by 3.6%(P<0.05), 4.6%(P<0.05), 2.7% and 1.4% respectively; The digestibility of hemicellulose was increased by 3.6%(P<0.05), 5.2%(P<0.05), 2.0% and 1.2% respectively; The apparent digestibility of calcium was increased by 12.2%(P<0.05), 14.3%(P<0.05), 8.2 and-4.1% respectively; Apparent digestibility of phosphorus increased by 16.7%(P<0.05), 17%(P<0.0 5), 8.3% and 4.2% respectively; Nitrogen retention increased by 30.5%(P<0.05), 39.6%(P<0.01), 14.2% and 8.2% respectively; Calcium retention increased by 5.3%(P<0.05), 7.5%(P<0.05), 4.0% and 0.3% respectively; Phosphorus retention increased by 18.6%(P<0.05), 24%(P<0.05), 9.7% and 5.3% respectively. The experiment concluded, The optimal AOT concentration of rumen fermentation was 0.20 mg/ml; To improve the feed intake of sheep feed intake, digestibility and nitrogen retention of the best AOT feeding dose of 1.2 g/kg diet; However, there was no significant difference at the administered range between 0.8 and 2.0 g/kg in voluntary intake and etc. indicating, a wider dose range for the beneficial administration of AOT.
