The experiment selected five male Small Tail Han sheep, at the age of about1.5years old, with a permanent fistula in theirs rumens, and were divided into5groups, supplemented with lysine-HCl at the dose of0%(Group1),0.4%(Group3) and0.8%(Group5) in diets, and were supplemented with urea at the dose of0.13%(Group2) and0.26%(Group4) in diets respectively. Every group labeled as0%(Group1),0.4%-lysine(Group3),0.8%-lysine(Group5),0.13%-urea(Group2, at the same level of Nitrogen with Group3) and0.26%-urea(Group4, at the same level of Nitrogen with Group5).5×5Latin square design was used in the experiment to study the effect of dietary lysine supplementation on the voluntary intake, the ruminal microflora, the digestion and metabolism of the rumen and whole body in Small Tail Han sheep. The results showed that the voluntary intake of dry matter of five groups were1625.4±108.2,1646.9±127.4(P>0.05).1714.1±107.1(P>0.05),1701.7±85.3(P>0.05) and1817.3±90.2(P<0.05) g/sheep/d respectively; The total number of rumen bacteria were102.21±9.91,105.85±10.95(P>0.05),121.51±11.00(P<0.01),107.76±12.41(P>0.05) and126.63±13.72(P<0.01)×109/ml respectively, the number of rumen species in bacteria were91.41±9.21,94.19±9.30(P>0.05),110.76±9.97(P<0.01),96.52±9.80(P<0.05) and118.92±7.96(P<0.01)×10g/ml respectively; The total number of rumen protozoa were8.31±1.17,8.62±1.32(P>0.05),8.70±1.23(P>0.05),8.59±1.28(P>0.05)and8.79±1.24(P>0.05)×105/ml respectively; The total number of rumen fungi DNA copies were9.19±2.17,9.31±2.35(P>0.05),10.38±3.35(P>0.05),9.64±3.22(P>0.05) and9.66±3.30(P>0.05)×107copies/ml respectively. The total VFA were104.00±19.57,103.57±15.16(P>0.05),113.95±13.87(P<0.05),111.88±14.54(P>0.05) and116.68±14.35(P<0.01) mmol/L respectively; the acetic acid were76.30±15.05,75.90±11.92(P>0.05).81.96±10.72(P>0.05),80.90±10.70(P>0.05) and83.18±9.29(P<0.05) mmol/L respectively; the metacetonic acid were15.14±2.89,15.53±2.35(P>0.05),17.82±2.48(P<0.01),17.69±3.21(P<0.01) and18.24±3.41(P<0.01) mmol/L respectively; the butyric acid were10.87±2.69,10.45±2.63(P>0.05),12.31±3.08(P>0.05),10.95±2.34(P>0.05) and13.05±3.32(P<0.01) mmol/L respectively. The concentration of total NH3-N were23.95±10.86,25.37±12.27(P>0.05),25.00±12.40(P>0.05),24.25±12.22(P>0.05) and25.27±11.74(P>0.05)mg/100ml respectively. The apparent digestibility of organic matter and crude protein were62.78±2.94%,64.21±3.71%(P>0.05),66.74±3.54%(P>0.05),66.75±3.53%(P>0.05) and69.02±4.09%(P<0.05);55.63±3.66%;56.48±6.30%(P>0.05),59.57±2.63%(P>0.05),61.06±3.41%(P>0.05) and65.89±1.38%(P<0.01) respectively. The nitrogen retention of the volume were2.81±0.79,3.92±0.96(P>0.05),4.15±1.04(P>0.05),4.52±0.50(P<0.05) and5.95±0.69(P<0.01) g/sheep/d respectively. It is concluded that supplementation of untreated lysine can increased the voluntary intake; the total VFA, the acetic acid, the metacetonic acid and the butyric acid; and the total number of rumen bacteria, the number of rumen species especially; the apparent digestibility of organic matter and crude protein; and the nitrogen retention of the volume. But the total number of protozoa and rumen fungi DNA copies were not promoted. And the effects by supplementation of untreated lysine were better than supplementation of urea. The conclusions results from the above:the total number of rumen bacteria at the adult sheep could be increased by supplementation of untreated lysine, therefore the ability of the digestion and metabolism of the rumen, and the nutritional status of the sheep can be significantly improved.
