Three year-old ‘Kyoho' grape under organic substrate cultivation and conventional cultivation was used as materials. To investigate the effect on ‘Kyoho'grape fruit quality, anthocyanin content in grape skins and expression of anthocyanin biosynthesis genes under organic substrate cultivation. Have a comparative analysis on phenological period and biochemical characteristics of ‘Kyoho' grape. And the fruit diameter, fruit weight, total soluble solids(TSS) content, titrateble acid(TA)content, TSS/TA and Vc content in different periods of grape were determined and free amino acid content and fruit texture in the mature stage were examined. The anthocyanin content in grape skins and expression of anthocyanin biosynthesis genes were analyzed by HPLC technology and q RT-PCR technology. The main results were as follows:(1) The period of injury flow, bud, flowering period, berry growth period, berries turn color and berries mature period of ‘Kyoho' grape under organic substrates cultivation were compared with the traditional cultivation about half a month in advance.The contents of chlorophyll and MDA in grape leaf were higher than that of control, and the activity of several enzymes, SOD, POD, CAT was lower than that of the control group.(2) The results indicated that the contents of total soluble solids(TSS), titrateble acid(TA), TSS/TA, Vc and soluble protein were significantly higher in ‘Kyoho' grapes fruit under organic substrates cultivation than those under conventional cultivation,and fruit diameter and weight were little different from the control group during the whole stage, but they were slightly reduced than those of control in the berries mature stage. This study showed that organic substrate cultivation could improve the quality of ‘Kyoho' grapes.(3) Nine kinds of anthocyanins under two kinds of cultivation conditions were detected, respectively. The results demonstrated that the percentage of each component of anthocyanins was changed under organic substrate cultivation, but had no influence on the component of anthocyanins.(4) The expression level of structural genes PAL, 4CL, CHS, CHI, F3 Hs etc and transcription factors Vlmyb A1-1, Vvmyb A1 etc was increased during the entire or part grape fruit coloring period under organic substrate cultivation. Correlation analysis showed that the transcriptional level of DFR, CHI, F3H1, Vvmyb A1 and Vlmyb A1-1 genes under organic substrate conditions was extremely significant or significant positive correlation with anthocyanin content, but that of PAL, F3H2 was negatively correlated and that of CHS1, DFR, CHS2, F3H1 and My B5 b was less correlated with the anthocyanin content. The transcriptional level of CHS1, DFR,CHS2, F3H1 and My B5 b genes under conventional cultivation conditions was extremely significant or significant positive correlation with anthocyanin content,but that of PAL, F3H2, 5GT was negatively correlated and that of F3'5'H, OMT was less correlated with anthocyanin content.
