A model of inflammation bowel disease in weaned pigs was established by intraperitoneal injection with LPS to study the effect of adding 0.2%lactobacillus delbrueckii which were added to the basal diets on the growth performance,intestinal morphology, serum biochemical parameters, cytokines secretion and the expression of inflammation-related gene and protein. The aim was to explore the possible anti-inflammatory action of lactobacillus delbrueckii which modulated IBD in weaned pigs, and providing scientific basis for the protective effect of lactobacillus delbrueckii on intestine.36 healthy weanling pigs (DuroeŚLandrace) were randomly assigned to 3 groups with 6 replicates per group and 2 pigs per replicate comp prising of control group,LPS control group and lactobacillus delbrueckii group, respectively. Control group and LPS control group were fed the basal diet, while lactobacillus delbrueckii group was fed the basal diet with 0.2% lactobacillus delbrueckii.The total test period lasted for 31 days with 3 days for adjustment. On the 22th,24th,26th and 28th day, pigs in LPS control group and lactobacillus delbrueckii group were intraperitoneally injected with 100?g/kg LPS, while pigs in the control group were intraperitoneally injected the same dose of normal saline. In the morning of the 1st,29th day, the body weight of pigs in each group was weighted. At the same time, the formal testing total feed intake and diarrhea of every pig was recorded. On the 29th day, one pig per each replicate was hollowly weighted and selected to collect blood sample from precaval vein, which was used for testing the serum biochemical parameters. At the end of experiment, one pig from each replicate was killed.The intestinal were separated into three segments to represent for jejunum, ileum and colon. These intestinal segments were in quadruplicate. Those were used for detecting intestinal morphology, the contents of cytokines, the expression of related gene and protein, respectively. The results were as follows:(1) Compared to control group, diet supplemented with lactobacillus delbrueckii could alleviate the decrease in ADG of weanling pigs after LPS challenge, and improved feed utilization ratio, but there was no significant difference(P>0.05). As for the diarrhea of weanling pigs, diet supplemented with lactobacillus delbrueckii could significantly reduced the increase in diarrhea rate after LPS challenge (P<0.05).(2) Compared to control group, diet supplemented with lactobacillus delbrueckii could improve the intestinal morphology and lower the score of intestinal tissue injury (P<0.05).(3) Compared to control group, diet supplemented with lactobacillus delbrueckii could inhibit the decrease of the serum IgG, IgA and IgM contents (P>0.05).(4) Compared to control group, diet supplemented with lactobacillus delbrueckii could significantly reduce the increase of inflammatory-related cytokines IL-1?, TNF-? and IFN-? contents in the jejunum, ileum and colon after LPS challenge (P<0.05). At the same time, diet supplemented with lactobacillus delbrueckii could improve the contents of ileal and colonic anti-inflammatory cytokine IL-10 in LPS control group, but the only significant difference was found in the colon (P<0.05). In addition, it failed to improve the contents of jejunal anti-inflammatory cytokine IL-10 in LPS control group.(5) Compared to control group, diet supplemented with lactobacillus delbrueckii could lower the elevation of inflammatory-related NF-?B protein expressionin all three intestinal segments, but there was no significant difference (P>0.05).(6) Compared to control group, the expression of jejuna and ileal HO-1 protein in LPS control group was enhanced, but there was no significant difference (P>0.05). However, there was a significant differce in the colon (P<0.05).Compared to LPS control group, diet supplemented with lactobacillus delbrueckii could significantly enhance the expression of jejuna HO-lprotein (P<0.05), but there was no significant differce in the ileum and colon (P>0.05).(7) Compared to control group, the abundance of NF-?B mRNA was significantly enhanced in all three intestinal segments of the LPS control group (P<0.05). Compared to LPS control group, diet supplemented with lactobacillus delbrueckii could significantly lower NF-?B mRNA abundance in all three intestinal segments (P<0.05).(8) Compared to control group, the abundance of HO-1 mRNA was decreased in all three intestinal segments of the LPS control group, moreover there was a significant difference in the jejunum and ileum (P<0.05). Compared to LPS control group, diet supplemented with lactobacillus delbrueckii could increase HO-1 mRNA abundance in all three intestinal segments, but there was a significant difference in the jejunum and colon (P<0.05).In conclusion, diet supplemented with lactobacillus delbrueckii had the alleviation of IBD in weaned pigs. It may be that lactobacillus delbrueckii reduced the expression and secretion of anti-inflammatory cytokines modulated by NF-?B signalling pathway, meanwhile lactobacillus delbrueckii increase the expression of HO-1.
