Scutellaria baicalensis Georgi,which is one of bulk medicinal herbs in China is perennial herb of Labiatae,Scutellaria L.It is one of authentic medicinal materials in Shanxi Province.Distributes the whole province.Scutellaria baicalensis Georgi’a root was recorded in the Chinese Pharmacopoeia.Baicalin,wogonoside,baicalein,Wogonin are the major medicinal ingredients with heat-clearing and damp-drying drug,purging fire for removing toxin,anti-pathogenic,anti-inflammatory,anti-free radical damage,anti-tumor,anti-allergy,antioxidant,postoperative analgesia,hepatoprotective,enhance immune function and regulate the central nervous system and other effects.With the improvement of recognition of Chinese herbal medicines,the market demand is increasing.Over-harvesting leds to the wild resources decline sharply.Cultural Radix Scutellariae exists unclear idioplasm,provenance mixed,quality unevenness and other issues.Naturalize and quantity producte of the wild resources is the urgent problem.In order to understand the genetic relationship of wild Scutellaria baicalensis from Shanxi,We used the RAPD technique to analyze twenty-nine varies of wild Scutellaria baicalensis and seven varies of test-tube plantlets of wild Scutellaria baicalensis from Shanxi.Established and optimized the rapid propagation system of fifteen varies wild Scutellaria baicalensis,and contrast the botanical character the test-tube plantlets.(1)The amplificate and cluster results show that:①18 primers of the twenty-nine varies of wild Scutellaria baicalensis amplificated 176 pieces,which are among 300 bp and 2 000 bp,including 167 polymorphic bands,each primer can amplificate polymorphic bands 4 to 18 pieces,each primer average amplificate 9.28 pieces polymorphic bands,and the polymorphic rate is 94.89%.Twenty-nine varies of wild Scutellaria baicalensis can be divided into six categories.②For the seven varies of test-tube plantlets 5 primers amplificated 20 pieces,which are among 500 bp and 2 000 bp,including 17 polymorphic bands,each primer can amplificate polymorphic bands 3 to 5 pieces,each primer average amplificate 3.4 pieces polymorphic bands,and the polymorphic rate is 85%.The cluster analysis result show that seven varies of test-tube plantlets can be divided into three categories.③The result show that there is a abundant genetic diversity among twenty-nine varies of wild Scutellaria baicalensis and seven varies of test-tube plantlets from Shanxi,and the genetic diversity is not relevant to the geographical distribution.(2)The culture and condition of the rapid propagation is followed:①MS+ 0.3 mg/L 6-BA is best for the bud proliferation,the light intensity need 6 600 lx,temperature is about 25℃(±1℃),light culture/darkness culture is 14 h/10 h,temperature difference is 3℃,the hnmidity should upon 60 7%,otherwise may led to the malformed seedling.②White with 0.5 mg/L DSC and 1.5 mg/L IBA is suit for rooting;1/2MS with0.5 mg/L DSC and 1.5 mg/L IBA is better for the root develop than White,the adventitious root cannot extend much that is good for the transplanting of the test-tube plant.③The optimization of the rapid propagation system find that 2 mg/L AgNO3 can promote the bud proliferation.④20 g/L~40 g/L sucrose is best for the test-tube plantlets.(3)Contrast the botanical character of fifteen varies test-tube plantlets,it show that the difference between different varies test-tube plantlets is large,but is small in the same variety,suggesting that most of the test-tube plantlets from Shanxi can express stably in progeny.So far,any system study on the rapid propagation and RAPD analysis of wild Scutellaria baicalensis from Shanxi haven’t been seen.The result provide experimental basis for genetic resources breeding,artificial cultivation and resources protection.
