Tibetan Mastiff. native to Qinghai-Tibet Plateau with desirable traits, is the only large-breed dog in China and belongs to the second class national protect animals. Previous studies mainly focused on genetics field, while less research reported the blood biochemical criterion of healthy Tibetan Mastiff in the province of Sichuan. China. This paper measured the blood biochemical criterions, together with serum glucose, calcium, phosphorus and its correlation hormone of healthy Tibetan Mastiff aged from 4 mouths to 7 years. All of the dog samples comed from Ganzi Tibetan Autonomous Prefecture, Ya'an City and Chengdu city. The whole results were divided into groups based on the sample source, age and sex of dogs, then analyzed statistically to study the difference of biochemical criterions and correlation hormone caused by age and sex. Because the serum glucose, calcium and phosphorus can be affected by eating, we choose eight Tibetan Mastiffs aged 2 to 4 years to harvest the serum samples at 30 min before eating and 30,60, 120 and 180 min after eating. Then we measured the content of insulin, glucagon, adiponetin, resistin, calcitonin, parathyroid hormone,1,25 dihydroxy-2 vitamin D3 in serum. All of the trial results are as follows.1. Researches on blood biochemical criterion of healthy Tibetan Mastiff of different age and sex. Experimental animals were divided into two groups according to sex,42 sample in male group, and 43 sample in female group; divided into 3 groups according to age, respectively 28 sample in I group (aged 0 to 2),33 sample in II group (2 to 4 years old) and 24 sample in III group (4 to 7 years old). The results show, the content of 1,25 dihydroxy-2 vitamin D3 of female is higher than that of male (P<0.05), while other content of biochemical indicators had no significant difference between male and female (P> 0.05). The serum cholesterol of 4 to 7 years old age group is significantly higher than 0 to 4 years old age group (P<0.05), while there is no significant difference in 0 to 4 years old age group (P>0.05). The serum triglycerides, high-density lipoprotein cholesterol (hdl-c) and low density lipoprotein content in 4 to 7 years old age group is significantly higher than 0 to 2 years of age (P<0.05), while there is no significant difference in aged 0 to 4 group (P>0.05). Low density lipoprote has no significant difference in each age stage (P>0.05). As the growth of the age. serum alkaline phosphatase levels decreased significantly (P<0.05). Serum aspartate aminotransferase and alanine aminotransferase levels have no significant difference among all age groups (P>0.05). The serum total protein content of 2 to 7 years old age group is significantly higher than the group aged 0 to 2 years (P<0.05) and there is no significant difference in the group aged 2 to 7 years (P >0.05). The content of serum albumin, globulin and white ball has no significant difference among each age stage (P>0.05). The blood calcium and blood phosphorus content in group aged 0 to 2 is significantly higher than the group aged 2 to 7 years (P< 0.05) while there is no significant difference in group aged 2 to 7years (P>0.05). The content of serum calcium, parathyroid hormone, and 1.25 dihydroxy vitamin D3 levels has no significant difference among all the ages (P>0.05).2. The studies about the impact of eating on blood glucose, blood calcium, blood phosphorus and its related regulation hormone in the Tibetan mastiff. The levels of blood sugar and insulin in Tibetan mastiff are at a peak in 30 min after eating, significantly higher than these before eating (i><0.05), while they return to the previous levels after eating 120 min. Glucagon level reaches the lowest at 30 min after eating, significantly below the level before eating (P<0.05); then it presents a rising trend but significantly lower than that before eating (P<0.05). The levels of adiponectin and resistin are not significantly changed in any-point-time before or after eating (P>0.05). Through correlation analysis the content variation of blood sugar is significantly positively related to insulin (P<0.05). negative to glucagon (P<0.05), no significant correlation with adiponectin or resistin (P> 0.05). The content of blood calcium and blood phosphorus in Tibetan mastiff increases obviously after eating 30 min, and presents significant difference before eating (P<0.05). while it reaches the peak at 60 min after eating, then presents a downward trend and goes back to the level without eating after eating 120 min and 180 min. The ratio of calcium and phosphorus drops to the lowest at 60 min after eating, and presentes significant difference at other point-in-time (P<0.05). There is no significant difference among calcitonin. parathyroid hormone, and 1.25 dihydroxy vitamin D3 levels at various time points before and after eating (P>0.05). Through correlation analysis, the change of blood calcium content is significantly positively related to blood phosphorus (P<0.05). while the change of blood phosphorus content is significantly negative related to calcium phosphorus ratio (P<0.05). Blood calcium, blood phosphorus, calcitonin, parathyroid hormone and 1.25 dihydroxy vitamin D3 have no significant correlation each other (P>0.05).
