Hydrangea is a deciduous shrub in the genus hydrangea.The excellent Hydrangea macrophylla ’Endless Summer’ is a variety of hydrangea,its flowering period is 10~12weeks longer than that of ordinary hydrangea,and its cold resistance is stronger than that of ordinary hydrangea,and it can also bloom normally in low temperature environment.In order to establish the rapid propagation system of Hydrangea macrophylla ’Endless Summer’,the tissue culture of Hydrangea macrophylla ’Endless Summer’ was studied by stem induction from bud and stem induction of callus.The results are as follows:1.The results of the study on inducing cluster buds from the annual stem of Hydrangea macrophylla ’Endless Summer’ were as follows: the best disinfection method for the explants was 75% alcohol for 30 s,followed by 0.1% mercuric oxide for 7min.Under this treatment,the mortality rate of stem was 3.3%,the contamination rate was13.3%,and the survival rate was 83.3%.The basic medium was MS medium,and the optimal combination of plant growth regulators was 6-BA2.0mg/L+KT0.5mg/L+IBA0.5mg/L,and the induction rate was 81.7%.KT had a significant effect on the induction of multiple shoots.The best combination of plant growth regulators was 6-BA 3.0 mg/L + IBA0.3mg/L,and the proliferation coefficient was 18.2.The results of callus induction from annual stem of Hydrangea macrophylla’Endless Summer’ were as follows: MS medium was the basic medium,and light culture was more conducive to callus formation;The best combination of plant growth regulators was 6-Ba 1.0mg/L + 2,4-D2.0 mg/L,and the induction rate was 100%.Compared with NAA,the induction effect of 2,4-D was better.The best combination of plant growth regulators for callus proliferation was 6-Ba 0.5 mg/L+2,4-D1.5mg/L,with a proliferation rate of 8.0.The best combination of plant growth regulators was 6-ba 2.5mg/L+NAA0.3mg/L,the callus differentiation rate reached 81.9%,and the highest average number of differentiated buds was 1.5.3.The results of the study on inducing the rooting of the adventitious shoots of Hydrangea macrophylla ’Endless summer’ were as follows: when the basic medium was MS and the plant growth regulator was 0.3mg/LIBA,the rooting rate could reach 93.3%.4.The results showed that when the seedlings were transplanted,the mixture of peat soil: vermiculite: perlite =2:2:1 was used,the survival and growth effect of sterile seedlings was the best,and the survival rate of seedlings was up to 90.0%.5.The results of the whole experiment showed that the best way to establish the rapid propagation system of Hydrangea macrophylla ’Endless Summer’ was to induce multiple shoots from annual stem segments.
