The breeding in chicken in our country is thought as traditional,that have long history more than thousands of years,With advancement in breeding techniques and social needs,the values and norms of intensive farming had become more significantly applied.But the problem of rising incidence rate is also under consideration and be ignored at the same time.Spleen is the biggest peripheral immune organs of chicken,however,up to now,few studies emphasize and interpret its function.The present study separated peripheral blood lymphocytes and labeled with 5,6-carboxy fluoresceindiacetate succinimidyl ester,CFSE,which is based on the researches of structural characteristics about white pulp in chicken spleen.The main prospective of present study was to track the process of lymphocytes homing in chicken spleen,discussing the meaning and providing theoretical evidences for the use in preventive medicine of chicken diseases.The following are specific research contents and results:1.The structure characteristics of splenic white pulp in chickenSpleen is major immune organs in chicken,so present study conducted to reveal the microscopic structure targeting on the observation of ellipsoid and penicilliform capillaries,moreover,we tried to interpret the relativity between the immune response and the microscopic structure of spleen.With the observation of light microscope,we observed that the endothelium of ellipsoid arteriole and penicillus is cuical-columnar endothelium,and the cell's junction between endothelium are not tight,immune cells were also observer in ellipsoid.We inferred that the penicillus may be an important location of chicken lymphocyte homing in spleen.2.Isolation and CFSE marking of lymphocytes in peripheral bloodWith advancement in technology,the methods of cell tracing has increased and CFSE method is better to trace cell in vivo,gradient centrifugation and CFSE fluorescence were applied in marking of living peripheral blood lymphocytes.We found that chicken peripheral blood lymphocytes were efficaciously isolated by Human Lymphoprep,marked by CFSE,the peripheral blood lymphocytes were viable and morphological intact,green fluorescent of marked cell was observed in fluorescence microscope,marking positive rate is 98%that proved the applying of gradient centrifugation in cell isolation is viable and efficacious and the experimental CFSE fluorescence method is a stable cell tracing method.3.Homing-tracing-relevant research of lymphocytes in chicken spleenAfter the injection of marked lymphocytes,some of these cells were distributed in red pulp while some were at the Peripheral Ellipsoid Lymphatic Sheath(PELS)in the site of red pulp bordering on white pulp.In addition,a small amount of lymphocytes were observed in the centre of white pulp ellipsoid.It's indicated that marked cell could entred the white pulp of spleen,there really exists lymphocyte homing in chicken spleen,and it's closely related to the ellipsoid,which is located the on outer side of sheath capillary.
