Agaricus biaporus is the most widely cultivated of edible fungi in the world.Casing soil layer is very important for the cultivation of Agaricus biaporus.This study was focused on three aspects:a)the effects of different casing soils on the growth,yield and quality of Agaricus bisporus;b)the change of microbial population and DGGE map of casing soil in different stage,to analyzing the cultured bacterial population;c)screening bacterias which promote pining of Agaricus bisporus.The major results were summarized as follows:1 The properties of different casing soilsPeat,due to its high porosity and moisture holding capacity,as well as low volume-weight,will not get hardened in case of repeat water applying and have perfect physical structure.Therefore,it is a main casing soil for agaricus bisporus applied during industrialized production at foreign factories.The experiment designed casing soil types with different formulations and studied their physicochemical properties respectively,including determination of moisture holding capacity,volume-weight,the Max.porosity as well as electrical conductivity.The results turned out that:The physicochemical properties of casing soil with wormcast and perlite are more conducive to growth of agaricus bisporus.2 Influence of different casing soil formulations on growth and output of agaricus bisporusThe growth status and output of agaricus bisporus under different casing soil formulations were measured.The results turned out that:1)The casing soil with wormcast presents higher moisture holding capacity.Moreover,the growth status of agaricus bisporus mycelium is better with shorter mushroom formation duration.2)The higher wormcast content,the higher agaricus bisporus yield will be.Combining with physical properties,growth status of agaricus bisporus,costs and other factors,it is suggested to take vegetable plantation soil mixing with 50%wormcast as casing soil during actual production.3 Microbial community structure at different soil covering periodsBacteria account for the most proportion among microorganisms in casing soil.The amount of bacteria will increase along with the growth of agaricus bisporus sporocarp.After the primordium formation of sporocarp reaching to the upper limit,the amount will start to decrease.4 Microbial diversity analysis at different soil covering periodsThe number of stripes and brightness of bacteria DGGE spectrogram at different soil covering periods are different,which indicates that differences exist on bacterial population structures and quantity at different soil covering periods.With the growth of agaricus bisporus mycelium,the number of stripes and brightness will increase to certain extent,and reach to the upper limit at primordium formation;at mushroom tide interval,the number of bacteria population will decrease,which indicates that Bacterial growth can promote primordium formation of agaricus bisporus,and that bacterial populations and bacterial population,as well as agaricus bisporus mycelium growth and metabolism degree present positive correlation.
