In this experiment,the young shoots of Quercus mongolica were used as test materials,Shearing born this year in the middle upper branches as cuttings,Using a single hormone ABT 1.NAA,IB A and the IBA+NAA mixed hormones=1:1?V:V?,four kinds of plant growth regulator respectively take 200 mg·L-1,300 mg·L-1,500 mg·L-1 and 1000 mg·L-1 a concentration gradient,The low concentration was immersed for 2 h,4 h and 6 h and the high concentration was quick diped for 10 s and 30 s,both compared with water,To explore the influence of different kinds and concentrations of exogenous plant growth regulator on cutting propagation of Mongolian oak.By using the method of high performance liquid chromatography?HPLC?to determine the dynamic changes of endogenous indole acetic acid?IAA??abscisic acid?ABA??zeatin?ZT?and gibberellic acid?GA3?;At the same time,the changes of nutrient?soluble sugar,starch,protein,total nitrogen.C/N?were measured during rooting of cuttings,And morphological and anatomical observation of the basal part of cuttings during rooting,exploring the Rooting Mechanism of Softwood Cuttings of Quercus mongolica.The main results are as follows:1.The exogenous growth enhancers could effectively promote the development of callus of Quercus mongolica.The callus development of IBA1000 mg-L-1 and IBA + NAA1000 mg·L-1 for 10 s was better,and the callus weight could reach 0.5 g and 0.52 g,but can not effectively promote rooting,rooting rate was only 3.3%.2.In the process of rooting,endogenous hormones IAA and ZT could promote the development of callus and promote the formation of adventitious roots.ABA inhibited the formation of adventitious roots.The higher level of GA3 could inhibit the rooting of cuttings.The ratio of IAA/ABA and ZT/ABA was positively correlated with the development of callus,and the ratio of GA3/ABA was negatively correlated with the development of callus.3.In the process of rooting.the change trend of soluble sugar content in the cuttings was decreased-rising and decreasing;the contents of starch and soluble protein were slowly accumulated and then consumed rapidly in the adventitious root expression stage,and increased in the adventitious root elongation stage.In the early cuttings increased significantly,and then decreased and stabilized in the adventitious root elongation of the slow accumulation;C/N ratio is higher,the more conducive to rooting.4.In the process of rooting of Mongolian oak,the root primordial cells appeared about 15-20 days after cutting.and the adventitious roots began to break through the epidermis at 40 days after cutting.The root primordia originated from the skin or callus and belonged to the mixed rooting type.The surviving cuttings form a complete root system.
