| MITF gene can code micropHthalmia-associated transcription factor,KIT gene codes stem cell factor receptor.Most of the literature study their association with animal hair color and important roles that they play in the process of forming melanin.It has been reported that MITF gene can regulate PGC-1α(peroxisome proliferator receptor γcoactivator 1-α),PGC-1α can affect the muscle function,such as muscle endurance,conversion of muscle fiber types.The gene can interact with MAPK1,and the activation of MAPK signal pathway can further increase the protein content of skeletal muscle fibroblasts,muscle fiber length and cross-sectional diameter,so it is speculated that MITF gene has impact on growth and development of muscle,and KIT gene is the target gene of MITF gene,it can control bone marrow hematopoietic stem cell HSC,which plays an important role in the normal generation of blood cells,then the red blood cells can provide oxygen for muscle cells to support the growth and development of muscle cells,so we speculated KIT gene also has a certain impact on the the growth and development of muscle,which will affect the slaughter,meat quality directly,so MITF gene and KIT gene are speculated to affect the poultry slaughter,meat quality.However,there are few reports on the association between these two genes and animal slaughter and meat quality traits at home and abroad.In this experiment,the second generation of white Kaiya×Liancheng and Maya 、Peking duck were used as experimental animals.11 SNPs in the MITF gene and 8 SNPs in the KIT gene were analyzed by PCR-RFLP,and the results were analyzed.We want to discuss the possibility of MITF and KIT gene as candidate genes for duck slaughter and meat quality traits.The main results were as follows:1.MITF and KIT genes were analyzed in duck tissues by expression profile,results showed that MITF and KIT genes were expressed in duck heart,liver,spleen,lung,kidney,stomach,leg muscle and chest muscle.2.A total of more than 90 suspected SNPs of MITF gene were found by RNA-Seq.11 SNPs were choosed to conduct RFLP.Their genotypes were association with duck slaughter and meat quality traits and we found that :(1)Some sites were related to part of the meat quality traits: c.808002C> T,c.808890C> T,C820188T、T844468C and G846879 A were correlated with the diameter of leg muscle fibers;C808890T、T810554G、C820188T、G834108T and T844865 C were correlated with color of the muscle in duck leg or chest;G810251T、C810282T and G834108 T were associated with duck chest muscle or leg muscle pH;G810251T 、C810282T、C810754A、G834108T and T844865 C were correlated with the chest muscle or leg muscle water loss;C810754A and G834108 T were correlated with the chest or leg muscle cooked meat rate;G834108T and T844468 C were correlated with the leg muscle or chest muscle broken coefficient;(2)Some sites were related to part of the slaughter traits: C810754 A and G834108 T were correlated with the slaughter rate;T810554G and T844468 C were correlated with the heart index;C810754A was correlated with the liver weight,liver index and and abdominal fat、sebum;3.The SNPs found in the duck KIT gene before were analyzed by RFLP in the resource group.The association analysis between genotype and duck slaughter and meat quality showed that:(1)Some sites were related to part of the meat quality traits: T5168C、A1238C、G1064C、A6578G、A1221T、A5557G and C1243 T were correlated with the duck leg muscle or chest muscle color;G1064C was correlated with the chest muscle or leg muscle water loss;A6578G was correlated with the chest muscle cooked meat rate;T5168C and A1221 T were correlated with the leg muscle or chest muscle broken coefficient;(2)Some sites were related to part of the slaughter traits: T5032 C and A1221 T were correlated with the slaughter rate;G1064C was correlated with the heart index,T5168C、A1221T、A5557G and C1243 Twere correlated with the abdominal fat or sebum;In conclusion,the SNP of MITF gene and KIT gene are associated with duck slaughter and meat quality.It is speculated that MITF gene and KIT gene can be used as candidate genes for duck slaughter and meat quality traits. |
