In this study,the genetic analysis of Flammulina velutipes"Jiangshan No.1" was carried out,the selection of nutrient elements,the optimization of liquid spawn and culture conditions,and the comparison experiments between liquid spawn and conventional solid strains were completed.First,the genetic diversity of "Jiangshan No.1" and the domestic variety of Flammulina velutipes were analyzed by cloning the rDNA-ITS sequence of Flammulina veluipes"Jiangshan No.1" strain and comparing with the ITS sequences of Flammulina velutipes cultivars in different regions of China.The carbon monoxide,carbon and nitrogen ratios and the constant elements,trace elements and vitamins needed for the growth of Flammulina velutipes"Jiangshan No.1" were screened out by single factor test,and the three kinds of preservatives were screened out.And the L9(34)type orthogonal experiment was used to further optimize the content of each component of the culture medium,and a nutrient formula suitable for liquid fermentation of "Jiangshan No.1" strain was obtained.The culture temperature,culture time,inoculation amount,bottling volume,shaking speed and initial pH value were optimized by single factor test.And finally obtained the best conditions of liquid spawn culture of the"Jiangshan No.1" strain,and then made a preliminary application.in the Shaxian Jinnong Industry Co.,Ltd.The results showed that liquid fermentation strains had obvious advantages compared with traditional solid strains.The aim of this study is to provide some reference for the cultivation and popularization of "Jiangshan No.1" liquid spawn.The main findings are as follows:(1)There were some differences between the strains of "Jiangshan No.1" and the domestic varieties of Flammulina velutipes,and the average genetic distance from other cultivars was 0.008,and the maximum genetic distance was 0.017.(2)Jiangshan No.1 nutrient elements:the most suitable carbon source for the flour,followed by corn flour;the best nitrogen source for the soybean meal,followed by yeast powder;hyphae in carbon and nitrogen ratio of 30:1 medium growth rate of the fastest;Potassium,phosphorus,calcium and magnesium ions in the constant elements can significantly affect the growth of Flammulina velutipes hyphae.Among them,potassium ions are most affected by the mycelial growth of Flammulina velutipes;the zinc and iron ions in the trace elements promote the mycelial growth The effects of six kinds of vitamins(biotin,riboflavin,folic acid,ascorbic acid,thiamine,niacin)on the growth of mycelium of Flammulina velutipes were not significant.The concentration of sodium diacetate in the accelerator was significantly promoted Mycelium growth.(3)After optimization by orthogonal experiment,the best nutritional formula was:corn flour 2%,soy flour 0.1%,yeast powder 0.1%,potassium dihydrogen phosphate 0.15%,sodium diacetate 0.02%.(4)The suiTab.culture conditions were as follows:temperature 25 ?,bottle volume 1OOmL(250 mL bottle of capacity),inoculation amount 10%,shaking speed 160 r/min,culture period 6 d,pH 6.0.(5)Comparison of liquid spawn and solid strains of fruiting strains:Compared with the solid strains,the germination time of the mycelium of the liquid strain was about 2 days ahead of the time,the mycelial bag time was about 10 days ahead of time,9 d or so.The average yield and biological efficiency of the fruiting bodies were better than those of the solid species.
