Pyrethrum cinerariifolium Trey.is a pesticide that has a good insecticidal activity and is less susceptible to drug resistance.The effective ingredient pyrethrin is mainly extracted from the flower of the P.cinerariifolium Trey.Therefore,the yield is greatly affected by the natural conditions,and the pyrethrin produced by the tissue culture has good application value.In this study,Agrobacterium tumefaciens LBA4404 and C58 were used to induce sterile shoots and calli from pyrethroids to generate adventitious roots and crown gall tissues.The hairy roots were treated with fungal elicitors to obtain high-yield P.cinerariifolium Trey.production system.The effects of major elemental concentration in the H medium on the synthesis of tripterine were studied by using Tripterygium wilfordii Hook.f.adventitious roots.The main results were as follows:1.The adventitious roots of pyrethrum could be obtained by induction of Agrobacterium tumefaciens LBA4404 without crown gall tissue and its viability was good and could grow in solid culture medium and liquid medium,and there was no sign of degradation.Which failed to detect pyrethrin,but could be detected in the treatment of Alzheimer's disease effects of the role of galanthamine,also had the value of industrial production.2.The use of P.cinerariifolium Trey.root hair for double conversion could be carried out,When the concentration of acetosyringone was 100 ?mol / L,the highest induction rate of double transformation was 85%.Negative pressure could decrease the induction rate of double transformation of pyrethrum.The induction rate of double transformation of A.tumefaciens C58 was higher than that of wild A.tumefaciens;ultrasonic treatment had no effect on the induction rate of double transformation.A.tumefaciens C58 was used to induce P.cinerariifolium Trey.crown gall cells.Crown gall tissue with white color could use suspension cell culture.3.The use of fungal elicitors on the pyrethrum hairy roots were treated and found that Botrytis cinerea,Colletotrichum gloeosporioides and Fusarium graminearum in the extraction of fungal elicitor could make the root of the pyrethrin content in the hair significantly increased,The fungal elicitors extracted from Phytophthora capsici could significantly increase the insecticidal activity of the hairy root extract to the three instar larvae of Culex pipiens pallens,when 24 h 1 mg/mL concentration could lead to more than half of its mortality.4.When in the H culture medium,N,P and K was 2.0-fold,Ca was 2.5-fold,Mg was 1.5-fold,there was the highest growth of adventitious roots of Tripterygium wilfordii Hook.f.;and when,N was 1.5-fold K was 2.0-fold,Ca,P and Mg were half,the content of tripterine was hightest;to make tripterine the highest output,the concentration of H the medium should be P and Ca of half,K of 2.0-fold,Mg of unchanged,N of 1.5-fold.Tripterine in the medium content was very small,for the adventitious root of 2.3%.
