Liriodendron sino-americanum belonging to Magnoliaceae Liriodendron.crossbred by Liiodendron chinense Sarg.and Liriodendron tulipifera L.is not only landscaping species,good ornamental tree,but also has good ecological benefits of afforestation tree species.Based on studies of Liriodendron sino-americanum cuttings during the growth of cuttings of physiological and biochemical under rooting agent and Agrobacterium rhizogenes treatment,trying to further improve the survival rate of Liriodendron sino-americanum cuttings,and theoretically rooting agent and Agrobacterium rhizogenes treatment of Liriodendron sino-americanum cuttings of physiological and biochemical characteristics and differences of rooting was going on.to provide theoretical basis and technical support for the Liriodendron sino-americanum working factory production.The results are as follows:(1)selection of Liriodendron sino-americanum cutting media and cuttingsLiriodendron sino-americanum cutting matrix selection formula 4(coconut husk:yellow soil:Perlite:peat =4:3:2:1)was most suitable for the survival of Liriodendron hybrid cuttings formed roots and cuttings,the average rooting rate was 53.3%.in the absence of Yellow soil,the matrix formulation of peat soil 2(coconut husk:Perlite =3:1)the average rooting rate is the lowest,only 16.7%.Liriodendron sino-americanum various types of cuttings can take root,carry bud cuttings average rooting rate was the highest,followed by the middle branch cuttings.The average rooting rate were 50.0%,46.7%,average shoot apical cuttings was the lowest,23.3%.(2)effect of Rooting agent on Liriodendron sino-americanum cuttingOrthogonal test of rooting agent ratio test,influence of Liriodendron sino-americanum cuttings rooting rate ranking ability is rooting agent types>rooting agent processing time>agent concentration.The optimum concentration at the concentration Zhifu rooting solution diluted 500 timesof hybrid liriodendrons cuttings soaking 3h.the best promoting effect.Rooting agent Zhifu,IBA treatment group,the control group of hybrid wood cuttings rooting rate and root growth of phenotypic characteristics are better than water treatment.Rooting agent Zhifu group had the best effect,the average rooting rate was 61.7%,the control was only 13.3%.In the process of Liriodendron sino-americanum cutting,leaf SOD activity,POD activity,proline content and IAA,ZR content increase and higher than that of the control.In the leaves of SOD,proline rich plant rooting agent response is more sensitive than IBA,peak earlier.Rooting agent Zhifu group of Liriodendron sino-americanum cuttings base ZR content was higher than IBA group,and IBA treated cuttings base IAA content at a high level;but always higher than that of control.Rooting agent Zhifu,activity of SOD,IBA in the treatment group of Liriodendron sino-americanum POD activity,proline content and IAA,ZR was significantly higher than that of control,MDA,ABA,GA3 was significantly lower than the control(3)effects of Agrobacterium rhizogenes on Liriodendron sino-americanumA4,ATCC11325 of Agrobacterium rhizogenes could significantly improve Liriodendron sino-americanum average rooting rate,promote the formation of adventitious roots of.A4 promote the best effect,the average rooting rate was 66.7%,the control group the average rooting rate was only 23.3%.and A4,ATCC 11325 of Agrobacterium rhizogenes could improve the antioxidant properties of leaf cuttings of hybrid tulip,enhance leaf photosynthesis,improve the root of endogenous hormones IAA,ZR content,reduced GA3,ABA content and increase the content of RNA in roots.Different treatment method for A4 of Agrobacterium rhizogenes 3(A4 bacteria liquid immersion liriodendrons cuttings after 30min,and then the optimal concentration ratio of rooting agent "Zhifu" for 3h)Liriodendron sino-americanum cuttings average rooting rate was the highest,the average rooting rate was 70%;the use of syringe draw A4 bacteria liquid acupuncture cuttings base the average rooting rate was only 38.3%..Application of A4 Agrobacterium rhizogenes and rooting agent Zhifu,using treatment 2(Agrobacterium rhizogenes A4 soaking liquid liriodendrons cuttings after cutting 30 min,every 15d watering the optimum concentration of rooting agent "Zhifu")the Liriodendron sino-americanum for cutting,the average rooting rate can be as high as 83.3%,not combined the use of rooting agent only with the control group A4 of Agrobacterium rhizogenes treatment the average rooting rate was 60.0%.and the rooting agent combination can also enhance Liriodendron sino-americanum cuttings average leaf area,leaf photosynthesis characteristics,enzyme activity.Enhance the root of endogenous hormones IAA,ZR content,reduced GA3,ABA content,and increased the root total RNA content.(4)Liriodendron sino-americanum biochemical changes and differences on the formation of root cuttingsIn the process of cutting,Liriodendron sino-americanum inserted SOD activity in ear leaf POD activity,proline content increased and decreased the content of MDA is conducive to the survival of Liriodendron sino-americanum cutting.The cuttings base soluble sugar content and soluble protein content increase and at a high level in favor of Liriodendron sino-americanum roots.cuttings base IAA.The content of ZR increased and decreased the content of GA3 is conducive to the formation of Liriodendron sino-americanum cuttings root;and ABA content increased in favor of Liriodendron sino-americanum enhance disease resistance,decreased is conducive to the formation of root cuttings.The leaf of Liriodendron sino-americanum photosynthesis,when leaf SOD activity,POD activity,proline content is high,MDA is low;is conducive to rooting.SOD activity,root POD activity,proline content and IAA,ZR,GA3,RNA content was high;the ABA content is low,there is conducive to the formation and growth of the AsLiriodendron sino-americanum roots of cuttings.
