Pleurotus eryngii,belongsto Eumycophyta,Basidimycetes,Agaricales,Pleurotaceae,Pleurotus.Because of both the almond taste and abalone flavor,it is also called almond abalone mushroom.Pleurotus eryngii has a very high nutritional value and medicinal value,known as the “mushroom king”,“scallop mushrooms”,“the reputation of the grasslands of Boletus edulis”."Spawn-burning",caused by cultivated at hightemperature,limited the yield and quality of the edible fungus in the past.In order to solve the problem of "spawn-burning",it is necessary to sdudy on the mechanism of increased respiratory rate at high temperature.The effects of culture temperature on growth rate,biomass yield,respiration rate,protein turnover and alternative pathway in Pleurotus eryngii are studied by means of oxygen electrode method and BCA method and so on in this paper.In order to lay a theoretical foundation and research ideas for edible fungi to solve the problem of "spawn-burning" in production.The results were summarized as follows:1.The strain Pleurotus eryngii ACCC52627 was provided by Agricultural Culture Collection of China.In order to study the effects of temperature on growth rate and biomass yield,the growth curves of cultures incubated at 15°C,25°C and 34°C was determined respectively.For stationary culture,the mycelial growth rate was the fastest for culture incubated at 25?,followed by that incubated at 15? and 34?.The mycelia biomass yield cultured stationarily at 15? was the highest,followed by that incubated at 25? and 34?.For shaking culture,the growth rate of mycelia incubated at 25?was the fastest,followed by that incubated at 34? and 15?.The mycelia biomass yield cultured shakingly at 15? was the highest,followed by that incubated at 25? and 34?.The results showed that the culture temperature of mycelium not only affected the growth rate of mycelium,but also influenced mycelia biomass yield,which is directly related to the later development of the fruiting body and its yield and quality.The higher the culture temperature,the lower the mycelia biomass,yield,and fruiting bodies yield.2.Respiratory O2 uptake rates of mycelia were measured by oxygen electrode method.The results showed that the total respiration rate increased with increasing growth temperature,and the respiration rate was the highest at 34?,which was significantly higher than that at 25? or 15?.3.By calculatd the respiratory costs of protein synthesis and protein turnover were calculated,it was found that the increase of respiratory rate was mainly caused by the increase of energy requirement in cell metabolism requirement,rather than simply due to the increase of respiratory enzyme activityunder high temperature stress.4.Alternative pathway inhibitor SHAM treated hyphae,concluded that under high temperature stress,the alternative respiration pathway and cytochrome pathway of mycelium were both increased,while the high temperature increased respiration rate was mainly caused by the increase of cytochrome pathway.Further verified that the increase of respiratory rate was mainly caused by the increase of energy requirement in cell metabolism requirement,rather than simply due to the increase of respiratory enzyme activity.
