Cucumber is an important melon vegetables,which has a wide range of cultivation.However,its weak roots,poor resistance resulted in the low yield of cucumber,and it is difficult to achieve annual production.With the development of grafting technology application in cucumber production,these problems have been greatly improved.Although,the varieties of cucumber rootstocks are not very rich.In order to screen out the suitable rootstock varieties for the region,Xintai sting was used as scion in this experiment,and 19 Chinese inbred pumpkins were used as rootstocks while the control was the seedling Xintai sting.The survival rate and agronomic traits of grafted seedling by different combinations of grafting were investigated.Then the clustering analysis of 19 pumpkin rootstocks was carried out by SPSS 19.0.We can conclude that grafting seedling of number 24 and 26 grown best,and No.29 and No.40 were in the middle level,however No.20 and No.35 were the worst.At the same time,the changes of grafting grafts were explored by measuring the related enzymes and grafting symbiotic affinity.The results are as follows:1.Rootstock selection.The scion was Xintaimici and the rootstock were 19 kinds of newly cultivated pumpkin,and respectively are 14,15,16,17,18,19,20,21,22,23,24,26,27,29,30,32,34,35,40.The seedling of Xintaimici was used as control,and then the survival rate was count after graft.After graft,all research was paid attention to until the grafted seedling grown up to three leaves with one core.The results showed that there were great significance in survival rates among 19 different combinations of grafting.The survival rates of No.24 and 26 pumpkins were above90% in average and others were at 70% to 90%.Twenty days after grafting,there were significant differences among 19 combinations of grafted seedlings in diameter of stem of scion,stock and the length of main vine and area of leaves.When the graft grown to three leaves with one core,the biomass,index of strong seedling and other agronomic traits were investigated and analysed by cluster analysis.The results showed that the grafting ability were best of 24 and 26,then 29 and 40 were in the middle level while 20 and 25 were rather poor.It can be concluded that the pumpkinstrains 24,26 can be used as the better rootstock varieties for the grafting.The research on the affinity mechanism was carried out in two aspects,respectively are the measurement of related enzymes and symbiotic affinity of graft seedling on the seventh day after grafting.The rootstocks in this experiment were pumpkins 20,24,26,29,35 and 40,which were selected in the first step.And the scion was Xintai mici.Finally,the grafting combinations of 26,29 and 40 were obtained by comparing the protective enzyme content of grafted seedlings and making correlation analysis of symbiotic affinity.This study provided an theoretical basis for the analysis of graft affinity by research on the mechanism and physiological and biochemical changes of cucumber and comparing the content of related substances in different grafting combinations.Also,the pumpkin species of 26 and 29 selected were suitable for greenhouse cultivation of cucumber in North China,which enlarge the application of pumpkins.
