The experiment studies the effects of nutrient restriction and compensation on blood indexes,ruminal and cecal growth in Mongolian lamb.Thirty male lambs,about 4-month-old,(average 20.70±3.51 kg body weight)were randomly allotted into control(C)and feed restricted(R)group.After nutrition restriction period(60 d),5 lambs were randomly selected in each group for slaughtered.Remaining 10 sheep in each group were re-separated into two groups and each group have 5 lambs.The groups(CH and RM)fed hey diet,the other groups(CM and RM)fed maize straw diet.The blood routine parameters,rumen and cecum epithelium morphology,proliferation,expansion,apoptosis,cell connection related protein gene expression were determined in the experiment.The results were as follow:1.Effects of Nutrient Restriction and Compensation on blood routine parameters in Mongolia lamb.Lamb venous blood was collected at the end of 60 d,90 d,and 120 d.The blood routine analysis with 12 indexes belonged to four categories was assayed by using blood routine auto analyzer.The results indicated that,after nutrient restriction,the number of WBC in RG group was significantly lower than in CG group(P<0.01),but all the other indicators show a declining trend.Compared with the control group(CH、CM),the PLT and WBCs of the restricted groups were significantly increased after 30 days of compensation and the HGB showed an increasing trend after 60 days compensation.During the whole compensation period,RBCs of formerly restricted group showed decreasing trend.2.Effects of Nutrient Restriction and Compensation on Morphology of Rumen and Cecum in Mongolian LambsBy tissue section,HE staining,and microscope measuring method,the effects of nutrition restriction and compensation on the morphological structure of mucosa epithelium was studied.The results showed that,after nutrient restriction,rumen papilla length of RG group was significantly shorter(P<0.01)and stratum corneum thickness was thinner than CG group.The thickness of cecal mucosa was significantly decreased(P<0.01),the Goblet cells were significantly reduced(P<0.01),Eosinophilic cells and white blood cells were significantly increased(P<0.05;P<0.01)in RG group as compared to CG group.After compensation,compared with the control group,RH and RM groups which rumen papillary height and thickness of stratum corneum increased(P>0.05),papilla width presented a downtrend.The number of erythrocytes in the cecum of RM group was significantly increased than that of CM group(P<0.01).the number of globular leukocytes in the cecum of RH group was significantly increased than that of CH group(P<0.01).Other blood indexes did not change significantly.3.Effects of Nutrient Restriction and Compensation on Proliferation,Expansion and Apoptosis of Rumen and Cecal Cells in Mongolian LambsThe total DNA content and total protein content of rumen and cecum were measured by UV spectrophotometer.The results showed that,after nutrient restriction,the number of rumen and cecal cells increased(P>0.05).after compensation,the rumen and cecum showed different changes:the number of rumen cells increased compared with the control group.the number of cecal cells was equal to the number of restricted period.Apoptotic cells in rumen and cecum were detected by means of TUNEL and microscope.The results showed that,after nutrient restriction,the number of apoptotic cells in rumen and cecal were significantly increased(P<0.01)in RG group.After compensation,the apoptosis rate of rumen and cecum cells returned to normal level(P>0.05).4.Effects of Nutrient Restriction and Compensation on Gene expression of Cell connection related protein in Rumen and Cecal Tissues of Mongolian LambsThe expression of Caveolin-1,Claudin-1 and Cx43 gene in rumen and cecum was analyzed by Real-time quantitative PCR.The results showed that the differences in the environment of the gastrointestinal tract.In the restrictional stage,the expression level of the restricted group in the cecum tissue was higher than that in the rumen tissue,and higher than the control group.In the compensation stage,the genes of rumen and cecal tissues have no significant differences by two kinds of compensation methods.
