Age at the first egg laying,egg production and egg weight,were important indicators to measure the egg production traits of laying geese.The egg laying performance is directly related to the selection and recruitment of the pre-hierarchical follicle and regulation of the related genes in geese.In this study,5 groups of geese were used as research objects in order to select the best combination of various production indexes.The differences among egg laying performance indexes were analyzed and compared by incubation test and analyzed the correlation between the indexes.The results were analyzed and compared base on the expression differences of PDGF and its receptor mRNA in different grades of pre-hierarchal follicles of geese at pre-laying and laying stages by using real-time PCR and western-blot techniques so as to provide experimental references and theoretical basis to improve egg production performance.The results obtained are as follows: 1.Statistic analysis of egg laying performance of different hybrid combinationsUnder the same nutrition,level environment and management,the egg production traits of 5 hybrid combinations were different.(1)Age at the first egg and the laying rate: group Ⅴ age at the first egg was the least at 239 days;group I age at the first egg was the earliest at 210 days;The egg laying rate showed that Ⅴ group was the highest and the lowest rate of group I the egg laying rate in group Ⅱ and Ⅲ higher than that of group Ⅰ 7.13% and 7.24% respectively.(2)average egg production:for the whole laying period,group Ⅴ the pure breed of Zi geese had the highest production performance about 55.8 eggs,group Ⅰ the pure breeding of Carlos geese had the lowest average production performance about 22.9 eggs.The total number of average eggs laid in group Ⅱ and Ⅲ was 36.6 and 36.8 respectively less than Zi geese however the number of eggs increased by 59.8% and 60.7%,respectively compared with that of Carlos geese.(3)Fertilization rate:the average fertilization rate of group Ⅳ was the highest,reaching 91.31%,and group Ⅱ was the lowest decreased by 25.54% compared with group Ⅳ was significant(P<0.05).The average fertilization rate in group Ⅲ and Ⅳ was better than that of group Ⅰ,Ⅱ and group Ⅴ.(4)Egg weight,egg shape index and birth weight:group Ⅱ and Ⅲ had the higher egg weight than group Ⅳ and Ⅴ increased 12.8%(about 33g)there was significant difference in egg shape index between the group Ⅱ,Ⅲ and Ⅳ(P <0.05);There was a strong positive correlation between egg weight and r=0.749(P < 0.01);There was a significant correlation between egg weight and egg shape index(P < 0.01);There was also a positive correlation between egg production rate and fertilization rate.2.Expression analysis differences of PDGF and its receptor in different grades of pre-hierarchical follicles of geese during the pre-laying stage.They were all expressed and translated into proteins of PDGF and its receptor mRNA in different grades of pre-hierarchical follicles of geese.Among them all,the expression of PE was the highest,and there was a significant difference compared with that of other grades of pre-hierarchical follicles(P < 0.05).There was no significant difference between MWF and LWF(P > 0.05).With the development of follicles,the expression of PDGF gene in mRNA and protein level gradually decreased,reached the lowest at SY and maintained at a certain level.The expression level of PDGFR gene in the mRNA continued to decrease and increased in LWF,and again decreased in SY.PDGFR protein was significantly reduced,and then gradually increased in MWF and LWF,and tend to be stable in SYF.3.Expression analysis differences of PDGF and its receptor in different grades of pre-hierarchical follicles of geese during the laying stage.The relative expression of PDGF and its receptor in different grades of pre-hierarchical follicles of geese during the laying stage were higher than that in pre-laying stage and the relative expression abundance was different.The expression of PDGF gene mRNA in SY was significantly lower than that of LWF,MWF and SWF(P < 0.05).There was no significant difference in the expression of SWF,MWF and LWF between the two groups(P > 0.05).The expression of PDGF protein in LWF was the lowest.SYF and MWF were significantly different from those of the other three grades of pre-hierarchical follicles,and there was no significant difference between SYF and MWF(P > 0.05).The expression level of PDGFR gene in mRNA gene showed a trend of decreasing and then increasing.The relative expression level of PDGFR protein was higher in PE in the process of development of the pre-hierarchical follicle.The relative expression of SYF was significantly reduced in SWF,and then increased and decreased in MWF and LWF.
