The effect of seasonal breeding and high broodiness has been seriously restricted the development of goose industry.Zi geese is one of the world high egg production geese breeds.The reason why it called “ Egg King” is because the geese is small and high egg production.Poultry is oviparous animals.The follicles can sustainable development,maturation and ovulation under the optimum ecological environment.Geese are fertilized when they reached sexual maturity.And poultry would not be form corpus luteum after ovulation.It is an ideal animal model.Zi geese were used as experimental animal.The aim of this study is the development of geese pre-hierarchical follicles.Neurotrophic factors(NTFs)family are part of proteins.The NTFs were composed of many neurotrophic factors.Brain-derived neurotrophie factor(BDNF),Nerve growth factor(NGF)and Neurotrophin-3 are the important member of this family.And they are the earliest and most popular neurotrophic factor.Previous studies have shown that NGF,NT-3,BDNF and its receptor TrkB can not only regulate the nerve cells,but also express in cardiovascular,endocrine,immune and reproductive systems.NGF related genes and its metabolite play an important role in the development of geese follicles.In order to understand NGF,the biological information of genes related to regulation and the expression tendency of related genes,this study was used high throughput sequencing.The transcriptome is the collection of three kinds of RNA.Total RNA was extracted from the 5 stages of pre-hierarchical follicles of geese,and sequenced by Illumine Hiseq2500 platform.This study obtained millions reads length of 150 bp.Goose is non-model organism as there is no suitable reference genome for goose.The good reads was assembled and annotated by GO and COG database.It can find out candidate genes related to reproduction,metabolism,growth rate and disease-resistant.It provides theoretical basis to molecular breeding and functional gene research.ERK pathway and genes related to this pathway were annotated by KEGG pathway.The FPKM value of each gene as the expression quantity.The RT-PCR and Western Blot were performed to analyze the expression of NT3,NGF,BDNF and its receptor TrkB.The results as follows: the four genes expressed in 5 stages of pre-hierarchical follicles.The expression of the four genes were highest in small white follicles.
