In this study, We use the pre-hierarchical follicles (primary follicles, small white follicles,middle white follicles, largest white follicles) of Zi geese and Kaluosi geese as material.RT-PCR (reverse transcription polymerase chain reaction) technique is used to quantify theexpressions of NGF mRNA and BDNF mRNA in pre-hierarchical follicles of different periodsand different geese breeds, and testing the change of follicles stimulating hormone(FSH),luteinizing hormone(LH),progesterone(PRL) and estrogen(E2) in two kinds of geeseblood which in different laying period simultaneously. Analysis of differentially expressed onNGF and BDNF of different reproductive performance of geese, and the expression of NGF andBDNF in the same species follicles at different stage, while exploring the relationship betweenthe content of FSH, LH, PRL and E2and the expression of NGF and BDNF which regulate theprocess of prehierarchical follicles. The study could be contributed to provide a theoretical basisfor further study of the regulatory mechanism of follicular development. Results as follows:1、The expression pattern of NGF and BDNF mRNAThe relative expression of NGF mRNA of same species at the same stage: During the peekperiod, the relative expression of NGF mRNA in primary follicles of Zi geese is significantlyhigher than that of other models (P<0.01), the large white follicles and small white follicles aresignificantly higher than that of middle white follicles (P<0.05).For Kaluosi geese,.And there isno significant difference among large white follicles, middle white follicles and small whitefollicles(P>0.05).For Zi geese, during the last stage of laying, primary follicles are verysignificantly higher than that of middle white follicles and small white follicles(P<0.01), thelarge white follicles are significantly higher than that of middle white follicles(P<0.05). Duringthe last stage of laying, the expression of NGF mRNA in large white follicles of Kaluosi geese isvery significantly higher than that of other models(P<0.01), the primary follicles are verysignificantly higher than that of middle white follicles and small white follicles(P<0.01).The relative expression of BDNF mRNA of same species at the same stage: During thepeek period, the relative expression of BDNF mRNA in primary follicles of Zi geese is verysignificantly higher than that of other models (P<0.01), the large white follicles are verysignificantly lower than that of other models (p<0.01).For Kaluosi geese, the primary folliclesare very significantly higher than that of other models (P<0.01). During the last stage of laying.,the relative expression of BDNF mRNA in primary follicles of Zi geese are very significantlyhigher than that of other models (P<0.01), The relative mRNA expression of BDNF in largewhite follicle was significantly lower than that in middle white follicle and small white follicles. The small white follicles are significantly higher than that of large white follicles and middlewhite follicles(P<0.05). For Kaluosi geese, the relative expression of BDNF mRNA in primaryfollicles is very significantly higher than that of other models (P<0.01).2、The expression of serum hormones, and the correlation and expression characteristic of NGFmRNA and BDNF mRNAThe correlation between the blood hormone and the pre-hierarchical follicles of Zi goose:theexpression of NGF mRNA in S follicles show significant positive correlation with theexpression of FSH in serum(P<0.05).The expression of NGF mRNA in S follicles showsignificant negative correlation with the expression of PRL(P<0.05).The expression of NGFmRNA in P follicles show significant negative correlation with the expression of LH and E2inserum(P<0.05). The expression of BDNF mRNA in M follicles show significant positivecorrelation with the expression of FSH and E2in serum (P<0.05). The expression of BDNFmRNA in M follicles show significant negative correlation with the expression of LH (P<0.05).The correlation between the blood hormone and the pre-hierarchical follicles of Kaluosi geese:The expression of NGF mRNA in S follicles show significant negative correlation with theexpression of PRL(P<0.05). The expression of NGF mRNA in P follicles shows very significantnegative correlation with the expression of LH and E2in serum and shows significant negativecorrelation with the expression of PRL (P<0.05).The expression of BDNF mRNA in M folliclesshows significant positive correlation with the expression of FSH and E2in serum (P<0.05) andshows significant negative correlation with the expression of LH and PRL in serum (P<0.05).
