The inferior performance of the geese laying is an impotant factor that causes the slowly development of goose industry in China,how to improve the performance is an important direction in the research of genetic breeding of geese.However,the development of goose follicular is closely related to the performance of goose egg laying,In recent years,a number of researchers with the aid of molecular biology techniques to investigate whether expression in some functional genes or differentially expressed in different periods of the follicle,and its direct or indirect effect on the properties of egg production.Therefore,this study combines traditional breeding methods with current molecular biology techniques to study the relationship between certain genes and egg production performance.In this study,the research Ⅰobject were four goose cross combinations which that(zi goose♂×zi goose♀Ⅱ),(Carlos goose♂× F1♀Ⅲ),(F1♂×zi goose♀ Ⅳ),(Carlos goose♂× Carlos goose♀),respectively.And through the hybridization to select the excellent hybrid combinations of egg performance indexes,and the relative expression levels of mRNA and protein of GRB2 and SOS in the ERK signaling pathway in geese pre-hierarchical follicles were using the real-time quantitative PCR and Western Blot.(1)The average egg production,average egg weight,egg shape index,fertilization rate and incubation rate of different hybrid combinations were determined by hybridization experiment.And the result showed that the average egg production has highest in group Ⅰwas 82.13±4.04,and it was significantly higher than other group(p<0.05);the average egg weight was 136.19±4.53 g in group Ⅰand significantly lower than other three hybrid combinations(p<0.05);the egg shape index were fron 1.45 to 1.55 were not significant difference for each group(p>0.05);the average fertility rate were 87.06%±8.62%,78.44%±7.84%,82.33%±9.66%,68.61%±2.99%,respectively,and the Ⅰ,Ⅱ,grouⅢ p were significantly higher than Ⅳ(p<0.05);the average incubation rate were 84.06%±9.87%,75.11%±6.55%,79.78%±7.74%,66.17%±3.4%,respectively,and the Ⅰ,Ⅱ,Ⅲgroup were significantly higher than Ⅳ(p<0.05);furthermore,there was a significant negative correlation between the egg weight and the fertilization rate and the incubation rate,and there was a significant positive correlation between the fertilization rate and the incubation rate.(2)The result of RT-qPCR showed that GRB2 and SOS gene were expressed at the levels of the mRNA and protein in ERK signaling pathway in pre-hierarchical follicles of geese,and the GRB2 and SOS genes had a highest expression in Primary follicles of geese that relative expression were 66.62±1.8 and 6.01±0.76,respectively,the lowest expression in middle white follicles was 31.61±0.82 and 0.12±0.04.The GRB2 and SOS had significant difference between the highest and lowest expression of mRNA(p<0.05).Protein expression analysis of GRB2 and SOS by Western Blot results showed that GRB2 protein had the highest expression in primary follicles was 2.33±0.02,and the lowest expression in middle white follicles was 1.25±0.02,there was a significant difference between the highest expression and lowest expression(p<0.05)of GRB2 protein,and there was no significant difference between large follicles and small yellow follicles(p>0.05).SOS protein had the highest expression in small yellow follicles was 2.137±0.02,and the lowest expression in primary follicles was 1.023±0.01 and significantly lower than that in other grades of follicles(p<0.05).(3)GRB2 and SOS are associated with follicle development,and regulate the development of follicles mainly by regulating the proliferation and differentiation of cells.
