Study on the Sex Ratio and GC-MS Identifiy of Adult Female Gonad Extract of Diaphania pyloalis WalkerDiaphania pyloalis Walker is also called oil insects,leaf worms,caterpillars,etc.,belonging to the Lepidoptera borer insects.The main parasitic plant of mulberry is mulberry,which is one of the main pests of mulberry.The larvae of the larvae are mainly harmful to the summer and autumn mulberry leaves.Mulberry borer control method is now mainly chemical pesticide control.However,due to the burst of mulberry and sericulture period is basically the same and pesticide colleagues will affect the development of silkworm and the use of long-term chemical pesticides mulberry borers have different degrees of resistance.New drugs and methods must be studied and explored.Pest control is mainly the use of natural enemies and sex pheromones.In this study,the composition of the gonadal extract of Sophora japonicus was studied.The main experimental contents and experimental results are as follows:The results showed that the natural proportion of females in the pupal period of Diaphania pyloalis Walker were 86.98% and 83.05%,and the female offspring of Single moth mating accounting for 82.91% in Zhenjiang yielding Mulberry garden and greenhouses mulberry garden from May to September.Besides,the Diaphania pyloalis Walker offspring by parthenogernesis was not found under natural conditions.GC-MS was employed to detect methylene chloride and n-hexane extracts of Diaphania pyloalis Walker Virgin moth gonad.Five compounds which has a certain similarity with known insect pheromone were discovered.They are 2-[[2-[(2-Eth)Met]Cyc]Met]Cyc,Aci,Met,Est,E2,Z15,Me14-18:OH,Z9,Z12,Z15-18:Aci:Iso:Est,Me6,Z8-14:OAc and Z9,Z12,Z15-18:Aci-4:Est,which could be used as a candidate for Diaphania pyloalis Walker sexual pheromone for biology identification.Diaphania pyloalis Walker in the form of mature larvae distributed in the mulberry fissure,insect hole or dead leaves in the winter.The mortality rate of winter mussel in northern China is high,but the mortality rate in the south is relatively low.The mechanism of dormancy induction and cold resistance of mulberry borers are lack of understanding.In order to understand the physiological and biochemical metabolic changes of the fifth instar larvae of Aristolochia manslata under low temperature induction.The expression of the gene and the content of metabolites in the low temperature were studied by transcriptome and metabolomics analysis.The results showed that the temperature of the mulberry Hibernation wintering mechanism provides clues.In the experiment,the larvae of the third instar larvae were divided into the following five treatments: the normal state mulberry moth was used as the control group(sample No.1),and the larvae were kept at 12 for 8h(sample 2)(Sample No.3),?16 days(sample 4),16 days and then at 4 ° C for 30 days(sample 5).40 larvae were sampled from each sample.1.Study on low-temperature induced transcriptomics of 5th instar larvae of Diaphania pyloalis Walker Five samples were extracted by RNA and sequenced.The number of Clean reads was 36736879-44374781,which was 85.84%-91.36% compared with the unigene reference sequence.From the Wein diagram and The KEGG enrichment scattergram of the differential gene showed that the sample No.3 was compared with the samples 1 and 5 and the No.1 sample.According to the results of the comparison,it was presumed that these changes may be related to the response or adaptation of the sleeping temperature to 4 ?related.The results of the above transcriptions can be concluded that the gene expression status of mulberry moth in the low temperature treatment at 12 ?has undergone significant changes.It is presumed that there may be significant differences in the gene expression between the 3 and 4 samples.The genes that are related to the mechanism of dormancy induced by hypothermia may be related to the low temperature adaptability of mulberry borers.2.Metabolomics of 5-year-old larvae of Diaphania pyloalis WalkerThe methanol-water mixed extract of 5-year-old larvae fed by low temperature was detected by UPLC-MS,Found 38 negative ion chromatographic peaks and 34 positive ion chromatographic peaks,a total of 72 small molecule compounds were obtained.The results showed that the content of 14 compounds increased with the prolongation of the induction time of low temperature.However,16 compounds decreased with the prolongation of low temperature time,suggesting that these substances may be highly correlated with the dormancy induction or dormancy process.According to the above results,it can be concluded that the larvae of the 3rd instar larvae of the 3rd instar larvae can cause changes in gene expression and metabolism and content of organic matter in the body at about 12 ?for about 8 days.These changes are related to the dormancy induction or low temperature adaptability may be closely related.
