For further research on Muscovy ducks abnormal behavior and better serving the livestock and poultry production and promotion of animal welfare,this experiment analyed Muscovy ducks abnormal behavior data and correlation analysis from the point of view of behavioral science and genetics.Through Media Record software to track Muscovy ducks behavior,and using the Observer XT software to define Muscovy ducks behavior,we developed Muscovy ducks behavior spectrum including the behaviors of exercise and rest?the behaviors of embellishing?the behaviors of alerting and aggression?the behaviors of abnormal?the behaviors of others.Based on a sample of 60 Muscovy ducks for experimental animals,we used DNA pool-PCR method,then direct sequencing method to screen SNP sites of NR3C2 gene and CRHBP gene,meanwhile used bioinformatics to analy the sequencing results.Through behavioral software,different behavior of Muscovy ducks can be divided into pecking anus Muscovy ducks group,pecked anus Muscovy ducks group,the stronger attacking tendency Muscovy ducks group and normal Muscovy ducks group.We used quantitative PCR(qPC R)to analy mRNA expression level of different groups of Muscovy ducks of NR3C2 and CRHBP gene in the brain,liver,kidney,chest,legs and bravery.The experiment results are as follows:1.The analysis results of behavior datas shown: the behaviors data of the behaviors of alerting and aggression and the behaviors of abnormal in pecking anus Muscovy ducks group,pecked anus Muscovy ducks group,the stronger attacking tendency Muscovy ducks group and normal Muscovy ducks group are obvious different,and the behaviors data in other groups are also obvious different.The frequency was positively or significant correlation with most of the behaviors,when making association analysis between different behaviors.2.Polymorphic sites and bioinformatics analysis results showed that the NR3C2 gene has 7 SNPs,respectively Intron2-G13928 T,Intron2-A13972 G,Intron2-T14336 C,Intron4-A34930 G,Intron4-T35157 G,Exon8 G198088 A and Exon9 G199134C;CRHBP gene has SNPs,respectively Exon3 G5805 A,Exon3 C5906 T,Exon7 A10747 G,Intron3 A5984 G,Intron3 T6019 G,Intron3 C6021 T,Intron3 G6035 A,Intron3 G6036 A,Intron7 C10506 T,Intron7 A10613 G,Intron8 G12097 A and Intron8-G12118 A.NR3C2 gene mutant before and after allele frequency estimation,mRNA secondary structure prediction result and secondary structure prediction result of NR3C2 protein had a significant difference,no obvious difference was found in tertiary structure prediction result of NR3C2 protein.CRHBP gene mutation estimation before and after allele frequency and mRNA secondary structure prediction result had a significant difference,secondary str ucture prediction result and tertiary structure prediction result of CRHBP protein had no obvious difference.3.The expression differences results shown: in four groups of Muscovy ducks,NR3C2 gene expression level has no obvious difference in the brain,liver and kidney tissues;In gallbladder tissue,NR3C2 gene exists significant differences in different groups of Muscovy ducks individuals,its highest expression level is normal Muscovy ducks,followed by pecked anus Muscovy ducks,stronger aggression Muscovy ducks,pecking anus Muscovy ducks is the minimum expression level.In four groups of Muscovy ducks,expression level difference of CRHBP genes in liver,kidney,chest,legs,and gallbladder tissue is obvious difference,and the expression level is extremely low;In the gallbladder tissue,expression level of CRHBP gene has obvious changes in different Muscovy ducks,and its highest expression level is normal Muscovy ducks,followed by pecked anus Muscovy ducks,stronger aggression Muscovy ducks,pecking anus Muscovy ducks is the minimum expression level.4.Correlation analysis of attack behavior and abnormal behavior results shown that the G199134 C site of NR3C2 gene and Muscovy ducks were no significant differences among indicators;A10747G site GG type of CRHBP gene and aggression traits index of Muscovy ducks was significantly correlated,shown that the genotype may be associated with aggression traits index of Muscovy ducks;G5805A and C5906 T sites and behavior traits of Muscovy ducks are no significant difference.
