With the development of animal husbandry,scientific research and production practice have scored new achievements.Semen diluent is broadly applied in artificial insemination.The capacity of seminal fluid could be expanded with the proper dilution,and it could increase the number of crossable female animals in one ejaculation of star male.On the other hand,it could be be benefit for the extension of stroge and transportation of seminal fluid,which make the star male more useable.For these reasons,artificial insemination is widely adopted and applied in more species in modern animal husbandry.Till now,there are many literatures about artificial insemination of broiler breeder with semen diluent.However,in the actual production process,the section of quality semen diluent,cycle of artificial insemination,and the usage of antibiotic in semen diluent are still not reported yet.In order to meet the demand of practical production and solve the novel problems in more poultry industry,this research mainly focused on the application of semen diluent in artificial insemination of Sanhuang broiler breeder.Test I:Screening of semen diluent of Sanhuang broiler breederContrast experiment of hatching egg were tested between storage seminal fluid and some commen semen diluents from home and abroad(diluted before use and preserve at room temperature).The optimized semen diluent was selected for popularization and application through statistical analysis.Sanhuang broiler breeder in four pheasantries with same breed,age,and count were obtained from some breeding chicken farm in Anhui Province.Artificial insemination was processed with storage seminal fluid and diluted storage seminal fluid with BPSE,Lake's,and Saline.The results showed that the average fertility rate of BPSE dilution group was lower than the control significantly(P<0.05).The average fertility rate of Lake's dilution group was higher than the control.The average fertility rate of Saline dilution group was lower than the control significantly(P<0.05).It can be concluded that the average fertility rate is higher when Lake's is adopted as the dilution.Moreover,the configuration of Lake's dilution is easy,which exhibits a good application prospects.Test ?:The investigation of cycle of artificial insemination of Sanhuang broiler breederThis section aimed at the extension of cycle of artificial insemination with the optimized semen diluent.Sanhuang broiler breeder in four pheasantries with same breed,age,and count were obtained from some breeding chicken farm in Anhui Province.The storage seminal fluid was diluted with Lake's dilution.Different cycle of artificial insemination of 5 days(control group),6 days,7 days,and 8 days were tested in the four pheasantries.The results showed that the average fertility rate of control group is the highest,while the 8 days group is the lowest.The difference between 6 days group and the control was not significant,while it is significant different between the other 2 groups and the control.Compared to the 6 days group,the different is significant with the 7 days and 8 days group.When the storaged seminal fluid was diluted with Lake's solution at the ratio of 1:1,there were no significant differences of average hatching egg fertilization rate between 5 days control group and 6 days group.It can be concluded that this dilution methods with Lake's solution could extend the artificial insemination cycle,which could save the labor and bring economic benefit for the company.Test ?:The application of antibiotic in semen diluentIn order to prevent salpingitis of chicken,the optimized concentration of gentamicin sulphate in semen diluent was investigated,and its influence of laying rate and hatchability was investigated as well.Sanhuang broiler breeder in four pheasantries with same breed,age,and count were obtained from some breeding chicken farm in Anhui Province.Samples were divided into four groups.In Lake's solution,0,5,10,20 mL of gentamicin sulphate was added,respectively.Then,artificial insemination was processed using the diluted storage seminal fluid with different concentration of antibiotic.The results showed that:(1)Compared to the control group,the incidence of salpingitis and death and culling rate exhibited a significant down trend in gentamicin sulphate applied group,while it is not significant among the three experimental group(P>0.05).(2)The average fertility rate exhibited no significant difference between 5%gentamicin group and the control,while it differentce is significant between experimental group(10%and 20%)and the control(P<0.05).(3)The difference of laying rate between the three experimental group(5%,10%,and 20%)and the control were all siginicant(P<0.05),while it is not significant among the three experimental group(P>0.05).To take both the economic benefits and the result into consideration,5 mL gentamicin sulphate in 100 mL Lake's dilution was adopted,which could prevent salpingitis without influence of average laying rate and fertility rate.Nowadays,Lake's dilution is applied in breeding chicken farm of 125 thousand poultry scale.It exhibited obvious advantages after appling 5 mL gentamicin sulphate in 100 mL Lake's dilution(dilute ration,1:1)and 6 days cycle of artificial insemination.The amount of male animals could be reduced by half,which could save the cost of almost 500 thousand.Moreover,the workload could be reduced,which will be benefit for the delicacy management.The third,the salpingitis could be prevented,and the average laying rate and fertility rate could be increased,which will improved the economic benefit obviously.
