In this experiment,Epinephelus malabaricus is experiment subjects,by the way of intraperitoneal injection research the influence of angelica polysaccharide on Epinephelus malabaricus non-specific immunity and immunity-related gene expression.Before the experiment,angelica first use absolute ethanol to remove impurities and decolorize.Then use the water alcohol precipitation method to get Angelica polysaccharides(CASP),Sevag method to remove protein,after dialysis obtain Angelica polysaccharide(ASP).Epinephelus malabaricus(68.14±1.75)g were randomly divided into nine groups,The group of injection of sterile saline as a control group,four were injected CASP,the other four were injected with ASP.The injection dose according to weight of fish was 37.5 mg/kg,75 mg/kg,150 mg/kg and 300 mg/kg.on the 3,7,10,14,21 and 28 d before and after injection continuous sampling.the intraperitoneal injection of CASP and ASP in a relatively short time(3-14d)can improve the grouper blood hemoglobin concentration,the number of NBT positive cells,plasma lysozyme activity,plasma A/G,plasma SOD activity and plasma C AT activity;decreased plasma MDA content;can reduce the infection of Edwardsiella tarda of cumulative mortality of grouper within seven days,improve immunologic potency.The low dose group of Angelica sinensis polysaccharide CASP(37.5-75 mg/kg)and the high dose group of ASP(75-150 mg/kg)have significant effect on the improvement of Epinephelus malabaricus non-specific immunity,relatively long duration.Sampling using real-time fluorescence quantitative PC R technology,related genes TLR-22,MyD88,TNF-?,TRIF and IRF3 expression.exploring the influence of intraperitoneal injection of Angelica Polysaccharide on grouper immune The results show that: the intraperitoneal injection of CASP and ASP can improve the expression of TLR-22,MyD88,TNF-?,TRIF and IRF3 in a certain time.Epinephelus malabaricus(78.14±1.75)g were randomly divided into 6 groups.The group of injection of sterile saline as a control group,the remaining 5 groups as experimental group,intraperitoneal injec tion of CASP,The injection dose according to weight of fish was 37.5 mg/kg?56.3 mg/kg?75 mg/kg?106 mg/kg and 150 mg/kg.on the 3,7,14 d before and after injection continuous sampling.The results show that: Injection of C hinese angelica polysaccharide CASP can significantly increase the proliferation of head kidney leukocytes,and can continue to 14 d,After injection of 3 d,53.6 mg/kg group,75 mg/kg group and 106 mg/kg group could significantly increase the proliferation of white blood cells,after injec tion of 7 and 14 d,all treatment groups could significantly increase the proliferation of white blood cells,the white blood cells proliferation ability of 37.5 mg/kg group is strongest.With the increase of injection time,head kidney leukocytes phagocyt ic activity gradually reduced.after injection of 3 and 7 d,all the treatment group can significantly improve the phagocytic activity of leukocytes phagocytic activity,37.5 mg/kg group was the highest.After injection of 14 d,37.5 mg/kg group,53.6 mg/kg group and 75 mg/kg group could significantly improve the activity of leukocytes phagocytic,37.5 mg/kg group was the highest.Results show that CASP can significantly improve the head kidney leukocytes phagocytic ability of grouper at low concentration,improve the immunity of grouper.Injection of C ASP can significantly improve the head kidney leukocyte respiratory burst of groupers.Comprehensive analysis shows that,the concentration of CASP is between 37.5 mg/kg and 75 mg/kg,each injection time can significantly improve the head kidney leukocyte respiratory burst of.And when the Angelica sinensis polysaccharide concentration was 75 mg/kg,at each measurement time,the head kidney leukocyte respiratory burst vitality of groupers were the highest.
