In this experiment,ascorbic acid was used as control,This study compared the antioxidant activity of Angelica sinensis polysaccharide(ASP)before and after protein removing.Angelica sinensis polysaccharide(ASP)was extracted with hot water and precipitated with ethanol.The ASP was obtained by removing protein from ASP by Sevag method.Ferricyanide reduction method,fenton method,DPPH method,and adjacent benzene three phenol autoxidation method were adopted to assay the reduction ability and free radical-scavenging activity in vitro.The results showed that Angelica sinensis polysaccharide(ASP)before and after protein removing exhibited significant reduction ability and free radical-scavenging activity(P<0.05).Within the same concentration,the reduction ability and free radical-scavenging activity of Angelica sinensis polysaccharide(ASP)before protein removing were higher than Angelica sinensis polysaccharide(ASP)after protein removing(P<0.05).However,the reduction ability and free radical-scavenging activity of Angelica sinensis polysaccharide(ASP)before and after protein removing were feebler than those of ascorbic acid.According to the above test results,Epinephelus malabaricus(89.61±2.08 g)as the test object,the strong antioxidant activity of Angelica sinensis polysaccharide(ASP)before protein removing according to different concentrations(0 mg/kg,2000 mg/kg,4000 mg/kg,6000 mg/kg,8000 mg/kg)were added to the diet,study in growth index,antioxidant index,antioxidant genes expression after 42 d and 84 d.Effects of dietary Angelica sinensis polysaccharide on growth index: the effect of dietary Angelica sinensis polysaccharide on the survival rate,weight gain rate,feed intake,feed coefficient,specific growth rate and fatness were not significant than control groupEffects of dietary Angelica sinensis polysaccharide on scavenging free radical ability:different concentrations of ASP could significantly improve the ability of superoxide anion radical and inhibit the hydroxyl radical of Epinephelus malabaricus,which was more obvious at 2000 mg/kg after 42 d and 84 d.Effects of dietary Angelica sinensis polysaccharide on antioxidant enzyme activity: 6000mg/kg of ASP in the diet had the best effect on T-AOC,Cu Zn-SOD,Mn-SOD,GSH-Px,GR,GSH,and decrease MDA content;8000 mg/kg group had the best effect on GST;in terms of improving CAT,the 2000 mg/kg group is the best.Effects of dietary Angelica sinensis polysaccharide on antioxidant genes expression in liver:feeding Angelica sinensis polysaccharide can significant increased Cu Zn-SOD,Mn-SOD,CAT,GSH-PX,GST,Trx,Prx gene expression.The GSH-PX,GST,Trx,Prx gene expression levels in the 6000mg/kg group were significantly higher than control group after 42 d and 84 d.Cu Zn-SOD 、 Mn-SOD gene expression first increased and then decreased by the ASP increased,Cu Zn-SOD,Mn-SOD gene expression in 6000mg/kg group was significantly higher than control group,Cu Zn-SOD gene expression in 2000,4000mg/kg compared with the control was group significantly decreased,Mn-SOD gene expression in 2000 mg/kg 、8000 mg/kg was significantly lower than control group after 42 d,in 84 d,4000 mg/kg group were significantly lower than control group,the other groups had no significant difference with control group.CAT gene expression in 6000 mg/kg was significantly higher than control group after 42 d,there was no significant difference between the treatment group and the control group after 84 d. |
