Soil ecosystem is one of the important factors affecting the quality and yield of tobacco.However,soil microbial diversity in its regulation plays an important role.This experiment sample traditional plate culture method and modern high-throughput sequencing technology from two aspects of culturable microorganisms and molecular ecology to explore the different fertilizer treatments on vega soil microbial diversity.The results were as follows:1.Through the plate culture method,we purification29 strains soil bacteria.Statistical analysis of different organic fertilizer processing soil samples of culturable bacteria diversity index.Cow dung compost and straw treatment can increase the number of culturable bacteria and richness of soil bacteria.Cow dung compost treatment effect of the most significant.Two compound fertilizer organic fertilizer A,organic fertilizer B and conventional fertilizer treatment on vega soil culturable bacteria number and types of influence is not obvious.2.Through Solexa genome analysis platform analysis of the 18 soil samples collected from 6 different vega processing soil fertilizer,in July,August,September.Received 1727576 original sequence,220697 OTU.OTU coverage of all samples are greater than 0.89,in combination with dilution curve,Shannon Wiener curve,the sequencing data can objectively reflect the extraction of soil samples in the microbial community structure and diversity characteristics.3.The Simpson index and Shannon index of 18 soil sample sequence-analysis.The soil applied organic fertilizer,which can influence the varying degrees of Simpson index and Shannon index of soil samples.Different organic fertilizer processing can influence the number of soil bacteria and species richness.Cow dung and fermented bean cake treatment on vega soil bacterial community richness most significantly,organic fertilizer A and organic fertilizer B the more conventional fertilizer treatment are showing A good soil bacteria abundance change effect,and straw organic fertilizer smaller influence on vega soil bacteria community richness.4.Use RDP classifier sequence classification of 18 samples status identification,these sequences belong to Acidobacteria,Actinobacteria,Bacteroidetes,Nitrospira,Proteobacteriaclasses etc.The Acidobacteria and Actinobacteria has the largest proportion of25.6% and 22.7%.The highest percentage of top 10 families.Sphingomonadaceae,Streptomycetaceae,Hyphomicrobiaceae,Bacillaceae,Streptococcaceae,Rubrobacteraceae,Nitrospiraceae,Nocardioidaceae,Koribacteraceae,Solibacteraceae.5.Different organic fertilizer processing significant effects on soil microbial florastructure.Cow dung compost and bean cake can increase the proportion of Nitrospira and Streptomyces.Organic fertilizer A can increase the proportion of Candidatus,Rhodoplanes and Streptomyces,but it reduce the proportion of Nitrospira and Rubrobacter of the Soil microbial.Organic fertilizer B and straw treatment compared with the conventional chemical fertilizer was not significant effect on advantage bacterium of the genus.6.Through PCo A,PCA and LDA difference contribution analysis,the microbial population structure similarity of 6 different fertilization treatmentin soil samples is higher,at73.45%,but there is still a significant difference between different processing.Consists of COG gap analysis,main microbial differences between the experimental samples in the signal transduction mechanisms and the carbon cycle and metabolism.
