Ovule and embryo culture technology is an important factor affecting the distant hybridization breeding.The varieties 'Fenyunu' of Paeonia lactiflora and 'Feng Dan Bai' of peony cultivar were used as parents to carry out the reciprocal cross in this paper.To compare the vitality of pollen grains during the storage of hybrid parents by measuring the vitality of stored pollen;To determine the optimal hybridization through statistic seed setting rate of reciprocal cross;The effects of different embryo ages and different medium on the growth were studied by train the ovule and embryo of reciprocal cross were cultured before abortion in vitro;The effects of sand storage and different GA3 treatments on seed germination and seedling growth of F1 generation were studied.The results show that:1.The pollen vitality of "Feng Dan Bai" and " Fenyunu " gradually decreased with the increase of time under the condition of storage for one year,The pollen activity of " Fenyunu " powder reached 58.6%,which was lower than that of "Feng Dan Bai" in the pollination for first days,which was about 10.5%.This shows that even in the absence of peony flowering period,but also the use of stored pollen to carry out distant hybridization2.It is of great significance to improve the seed setting rate of hybrid peony breeding,hybrid combination is the key to seed setting rate.In this experiment,the setting rate of orthogonal was 7%,and the seed setting rate of anti cross was as high as 100%,the seed setting rate of anti cross was significantly higher than the setting rate of orthogonal,This may be related to the growth potential and the strength of "Feng Dan Bai" has a certain relationship.3.From the beginning of ovule culture show: The effect of embryo age on ovule culture was obvious,The development of early?? 15 d?of ovule culture is not successful.At the age of 9 d and 10 d orthogonal ovule swelling rate is higher,the survival time is relatively long;At the age of 11 d,12 d,13 d anti cross ovule culture process in the early stage of rapid growth,cultivation of 60 d all turned green and the swelling rate was relatively high,However,all of them were gradually stopped growing after 60 d culture.The indexes of ovule growth were relatively good at MS+NAA0.5 mg/L +IBA0.2 mg/L or IBA0.5 mg/L+3% sucrose in the first 45 d after ovule culture of orthogonal;The indexes of ovule growth were relatively good at MS+NAA0.5 mg/L +IBA0.2 mg/L or IBA1.0 mg/L+3% sucrose in the first 60 d after ovule culture of anti cross.In the subculture,the positive and negative ovules began to stop growing and gradually died after 80 d culture.4.Embryo culture showed that: The differentiation and differentiation of embryos were also different in different culture media.With MS as the basic medium with different concentrations of 6-BA and NAA after hybrid early embryo normal growth medium were screened,consider the swelling rate and germination rate and uniformity is relatively better at MS+6-BA1.0mg/L+NAA0.1mg/L+3% sucrose in primary culture medium.The initial medium seedlings transferred to subculture medium after 30 d index statistics found: The optimal ratio of each index is MS+IBA0.2 mg/L+GA30.2 mg/L+6-BA0.2 mg/L+3% sucrose and MS+IBA 0.1 mg/L+GA30.3 mg/L+6-BA0.2 mg/L+3% sucrose.A better culture effect about root number and root length of a plurality and protrusions was relatively high at MS+IBA0.3 mg/L +NAA0.2 mg/L+3% sucrose after the statistics of 30 d.5.The seed germination of hybrid F1 results showed that: Sand storage and GA3 in advance of the hybrid seeds rooting period and the percentage of adorable rooting rate and root length ?3cm rate were higher.Dispose 2?300mg/L GA3?can obtain the highest rooting rate?88.75%?and root length ?3cm?22.08%?.The suitable root length had an important effect on breaking dormancy of Peony Seed.The study of GA3 hybrid seed treatment on root length ?3cm to rescind epicotyl dormancy was better,The each index consist of germination rate and pumping rate and uniformity of dispose 1?100mg/L GA3 for 2 h?was highest,catalytic seed germinate rapidly.
