Quality of traditional Chinese medicine in relation to the quality of seedling.Huaiqing Chrysanthemum is one of the top four in traditional Chinese Chrysanthemum.The producing areas of Huaiqing Chrysanthemum morifolium what mainly rely on cuttage propagation and root division propagation,with the use of auxin and carbendazim in grow seedlings,and the problem urgent to solve is to select the proper seedling treatments for raising the excellent seedlings of Huaiqing Chrysanthemum morifolium.The materials of this study come from Huaiqing Chrysanthemum,the effect of root treatments(half-cut,root division,whole-cut)and medicament treatments((CK),auxin,carbendazim,auxin+carbendazim)are designed to analyze by a comprehensive index of yield and quality of Huaiqing Chrysanthemum morifolium.The aim of this dissertation is to research the yield and main active ingredient contents of the Chrysanthemum morifolium in different harvest time and processing methods basing on the optimizing combination of seedling treatment.The results of this dissertation help to provide theoretical basis for the standardized seedling raising and processing technology of Chrysanthemum morifolium.The main results are as follows:1.In terms of stem diameter,the simultaneous use of auxin and carbendazim can provide more suitable conditions for the growth and development of plants,but there is no difference among all seedling treatments at last.In terms of plant height,it is higher in the whole cutting and root cutting treatments,and the plant height of the two root treatments is respectively higher than that of the half cutting treatment at the rate of 13% and 12%,at the simultaneous use of auxin and carbendazim.Meanwhile,The plant height of Huaiqing Chrysanthemum morifolium treated by root division treatment at the simultaneous use of carbendazim is respectively higher than that of the treatments of CK and auxin,11.9%?12.5%?2.There is little effect on grain weight per plant of Chrysanthemum morifolium between different medicament treatments,while root treatments have a big difference,and the half-cut root treatment is higher than root division 21%-32%.In addition,there is no difference on grain weight per plant of Huaiqing Chrysanthemum between different processing methods.3.The comparison of total flavonoids content of Huaiqing Chrysanthemum in different treatments:different root treatments have little effect on the total flavonoids content of Chrysanthemum morifolium Ramat,and the total flavonoids content of whole-cut root treatments is the highest under the condition of water(CK)and carbendazim treatments.The total flavonoids content of Huaiqing Chrysanthemum is higher under the condition of auxin medicament treatments,and is 113%,116% than that of carbendazim,auxin+carbendazim medicament treatments.In terms of the total flavonoids content in Huaiqing Chrysanthemum,there is no significant difference in different harvest time,and steaming baking treatment is higher than direct baking treatment 16%?24%?32% at the same period of harvest,respectively.4.The chlorogenic acid content of Huaiqing Chrysanthemum between different medicament and root treatments is 0.77%-1.05%,meeting the standards of the 2015 edition of Chinese Pharmacopoeia;the content of galuteolin between different medicament and root treatments is up to 0.10%-0.25%,meeting the standards of the2015 edition of Chinese Pharmacopoeia,while some treatments are as low as0.003%-0.079%;the content of 3,5-O-double caffeoylquinic acid of Huaiqing Chrysanthemum is up to 0.72%-1.07%,meeting the standards of the 2015 edition of Chinese Pharmacopoeia,while some treatments are as low as 0.01%-0.69%.Meanwhile,auxin can lead to a rise 15%-36% in the contents of chlorogenic acid and a rise 49%-59% in the contents of 3,5-O-double caffeoylquinic acid of Huaiqing Chrysanthemum compared to other medicaments,and the contents of chlorogenic acid and 3,5-O-double caffeoylquinic acid of Huaiqing Chrysanthemum are much higher under the condition of whole-cut and half-cut root treatments.5.There is some difference in the contents of chlorogenic acid,galuteolin and3,5-O-double caffeoylquinic acid of Huaiqing Chrysanthemum in different harvest time and processing methods.As for the content of chlorogenic acid in Huaiqing Chrysanthemum morifolium,the optimum harvest time is the later period of harvest,and the content of chlorogenic acid of the steaming and baking treatment is 113% ?118% ? 103% than that of direct baking according to harvest time,all treatments meet the standards of the 2015 edition of Chinese Pharmacopoeia;as for the content of galuteolin in Huaiqing Chrysanthemum morifolium,only November 2nd harvest and the steaming and baking treatment meets the standards of the 2015 edition of ChinesePharmacopoeia;in terms of the content of 3,5-O-double caffeoylquinic acid,different processing methods have no difference,and only November 2nd harvest meets the standards of the 2015 edition of Chinese Pharmacopoeia.
