There are 14 kinds of natural species of Hemerocallis,mainly distributed in Eurasia temperate and subtropical areas.Our country is day lilies with complete plant species of the genus and the distribution of the most widely countries,existing 11 for our country of origin,mainly concentrated in the south of Qinling subtropical regions.Since modern times,the breeding of plants of the genus day lilies at home and abroad,the development work,gardening breed new constantly,only the United States,there are tens of thousands of varieties are breeding.Entering the new century,our country gradually introduced many horticultural varieties from abroad,combined with the domestic independent breeding new varieties,wide variety,give the relevant identification work caused great trouble.Therefore,for day lilies gardening breed resource classification statistical work become very necessary.This experiment respectively from the morphological character,SSR molecular marker and ISSR molecular markers such as three methods for 50 day lilies classification research on plant material,laying the groundwork for plants of the genus day lilies classification work.Based on the experimental results are as follows:1.The biological character of the experiment selected 17 day lilies taxa of 50 day lilies material investigation,evaluation,and the results show that the index difference obvious seven biological quality characters,coefficient of variation range of 8.51% to 226.68%,the average of 60.98%.In terms of the characters of source,50 day lilies factors of plant material quality characters of stems mainly from design and color,fragrance and double.10 index of quantitative character,plant height,pedicels,and leaf length variation is more obvious,coefficient of variation were 44.12%,36.64%,33.90%,the average coefficient of variation is 24.10%.2.Through the R type of 44 clustering results showed that the biological character were selected for the experiment,there is a connection between each other,and independent of each other,the characteristics of the plants of the genus day lilies that can be used as a material properties;Q type cluster results showed that 44,between material when genetic distance is 6 to spread rapidly,and the two copies of the original species of day lilies in the genetic distance of 12 varieties was separated.When the genetic distance of 15 and 44 material is divided into four major categories.3.This experiment related to look for the plants of the genus day lilies in the NCBI microsatellite sequences,using SSR hunter software find out 100 SSR sequences,through Primer5.0 software to find out the microsatellite sequences primer design,design a total of 246 pairs of evaluation of higher,finally by comparison,filter,select one of the 100 pairs of primers are conducted PCR amplification,and finally the sifting has good polymorphic 20 pairs.Using Popgene32 software is analyzed,the results found that lead RY31 HemC203 and two pairs of primers on the material of the amplification effect is best,amplified out of the eight pieces respectively,polymorphism bands for eight,the polymorphism rate was 100%.Primers RY22 with primer RY34 amplification pieces at least,is only 2,polymorphism bands respectively 2,the polymorphism rate was 100%.20 pairs of primers for 50 material amplification out a total of 101 bands,each primer amplification all with 5.05 on average,including 92 polymorphic bands,polymorphism percentage is 91.10%.4.Through the SSR was carried out on the 50 material-PCR amplification,propagated the fragments through statistics,use Popgene32 and NTSYS software on 50 choose grass plant material for genetic distance and similarity coefficient computing statistics,calculation results show that the genetic distance between 0.0798 and 0.7794,while the similarity coefficient between 0.5510 and 0.8418.5.The use of article 17 primers for 50 day lilies material for ISSR-PCR amplification,using polyacrylamide gel electrophoresis to test the amplification of the product,by 17 primer amplification out 196 rules,including 189 polymorphic bands,polymorphism percentage is 96.43%,the average each primer amplification of article 11.53.Most of primers UBC811 amplification of banding,amplification bands 18,polymorphism bands 18,polymorphism is 100%;Primers UBC829 and UBC864 two primers amplification of banding,at least for the eight,polymorphism bands for eight,polymorphism is 100%.6.Through the study of the ISSR-PCR amplification of 50 materials,propagated the fragments through statistics,use Popgene32 and NTSYS software on 50 choose grass plant material for genetic distance and similarity coefficient computing statistics.Results showed that the genetic distance between 50 material between 0.0085 and 0.6794.While the similarity coefficient between 0.4920 and 0.6031.
