Tea is an important economic crop in China.Cuttage is the most important method of asexual reproduction.In order to further shorten the breeding cycle time of tea clone,improve the quality of them,explore the suitable materials and the appropriate endogenous and exogenous hormone levels in rooting and growth of tea tree,Camellia sinensis?L.?O.Kuntze cv.Taicha 12 was used as research material.The content and distribution of the related physiological and biochemical components were studied in this experiment.The effect of the pre-treatment of mother tree,the retention or removal of apical-shoot,the number of mature functional leaves,different soaking time of rooting agent and endogenous phytohormones on the rooting and aerial growth of tea cuttings were also explored.The main results were as follows:1.Soluble sugar and chlorophyll contents increased while the leaf became more mature.The net photosynthetic rate also raised with the increased maturity of leaf and the highest rate was the second mature functional leaf.With the increase of leaf maturity,the content of soluble protein decreased,and the apical shoot with one bud and tender leaf possessed the highest content.The second functional leaf possessed the highest contents of IAA and ABA,The content of ZR in the third functional leaf was highest,and the content of GA3 increased with the increase of leaf maturity.The content of IAA of the basal stem?S1S3?near the functional leaves of tea twig was significantly higher than S4.However,the content of ABA in S1 was significantly higher than that of S2S4.2.Spraying plant growth regulators such as DA-6 and foliar fertilizer on tea tree was beneficial to increase the chlorophyll content and net photosynthesis rate of leaves,which was beneficial to the formation of adventitious root and the growth of shoots and leaves.Spraying30ppm DA-6 was more beneficial to shoot growth.Spraying 2030ppm DA-6 was conducive to the root development.And a certain amount of foliar fertilizer had some auxiliary effects.3.The survival and rooting rate of cuttings with mature functional leaves were both100.00%,while the survival and rooting rate of single-bud cuttings with one or two tender leaf were only 5.00%and 23.33%respectively.The best materials for rooting were the cuttings with apical shoot and three mature functional leaves.Such cuttings possessed the fastest speed of rooting and had the maximum amount of roots.A large number of roots emerged from the base of stem segment within 10 days and the formation of secondary roots of 6070%tea cuttings only required 37 days.And the number of new matured functional leaves was also higher than others.However,the increased value of new height is less than the cuttings with one bud and two tender leaves.And the increased value of basal stem diameter is also smaller than cuttings with two functional leaves.Increased number of mature functional leaves was conducive to root growth and development,and it can also improve survival and rooting rate.The rooting and growth of cuttings whose top shoot was preserved was better than clones of which apical portion was removed.4.The content of IAA in the basal stem significantly increased after soaking the rooting agent,and the IAA/ABA and?IAA+ZR+GA3?/ABA values increased to about 1.0,which is conductive to rooting.During the first hour of soaking the rooting agent,the content of IAA rapidly increased and the IAA/ABA and?IAA+ZR+GA3?/ABA values also increased to about1.0 in the cuttings with one or two functional leaves and apical shoot.For other cuttings,the above three indicators reached the maximum value after soaking the rooting agent for four hours.F1-1,F2-1 soaking the rooting agent for 0.51h,F1-2 soaking the rooting agent for 4h,F2-2,F3-1 and F3-2 soaking the rooting agent for 34h,the content of IAA reached a high level,and IAA/ABA and?IAA+ZR+GA3?/ABA values can be increased to about 1.0.At the time of transplantation,the maximum values of the average root vigor,average rooting number and average increased value of basal stem diameter occurred in soaking the rooting agent within 02h.The other growth indicators of root and aerial parts performed better with the soaking rooting agent for four hours.
