Mechnasim Of Bovine Casein Glycomacropeptide (CGMP) Affecting Ulcerative Colitis Of Mice

Casein glycomacropeptide(CGMP)is a resource of bovine bioactive protein with potential for further exploration,which has been proved to have many bioactivities.CGMP has nutritional identity,as well as immune regulatory,improving intelligence,anti-microbes and anti-toxin,promoting probiotics and prevention for decayed tooth.Some diseases such as inflammatory bowel diseases can be caused by the turbulence of immunological homeostasis.Ulcerative colitis,as one of inflammatory bowel diseases,is a chronic relapsing disease of intestine with unknown causes.Appropriate nutrition preparation has been considered to be a long-term therapy for ulcerative colitis.Oxazolone was applied to set up mice model with ulcerative colitis.The effect of bovine casein glycomacropeptide on ulcerative colitis was investigated first,and the technologies of CBA and ELISA were applied to detect the content of IL-1?,IL-2,IL-4,IL-5,IFN-?,TNF-? and IL-10 in serum of mice with ulcerative colitis on the basis of our research on the cytokines of normal mice,and IHC was used to detect MUC2 of all the mice,then western blotting was used to detect the expression difference of MAPK p-p38 and NF-?B p65,in order to explore the mechnasim of bovine casein glycomacropeptide affecting ulcerative colitis of mice.Results: CGMP could decrease the diamine oxidase(DAO)activity in serum,myeloperoxidase(MPO)activity,malonaldehyde(MDA)content in colon,and the result of tissue morphologic of colon indicated that CGMP could keep integrity of epithelium.The effect of CGMP on IL-4 and IFN-? of normal mice suggested that CGMP could regulate Th1/Th2 imbalance.CGMP down-regulated IL-1?,IL-5,IFN-?,TNF-? and up-regulated IL-10 in the serum of mice with ulcerative colitis.The secretion of MUC2 was also promoted by CGMP.The signal passway of NF-?B could be inhibited significantly by CGMP.Although CGMP could not inhibit MAPK significantly,the trend of decrease was not ignored.Conclusion: CGMP has significant treatment for ulcerative colitis.The mechnasim of bovine casein glycomacropeptide affecting ulcerative colitis of mice is that CGMP could down-regulate IL-1?,IL-5,IFN-?,TNF-? and up-regulate IL-10 through inhabiting the activity of NF-?B and MAPK,and promote the secretion of MUC2.
