Experimental objective:Rabbits with steroid induced avascular necrosis of the femoral head model is established by use of drugs, the bone erosion capsule No.1 and No.2 to drugs in the process observed shares bones of CD34、 CYR61 and CTGF expression influence and shares of bone tissue damage and repair situation. In order to provide an effective experimental basis for the clinical treatment of steroid induced femoral head necrosis.Experimental methods:Effect of Bone erosion ning capsule on CD34、CYR61 and CTGF expression in rabbits with steroid induced avascular necrosis of the femoral head60 healthy adult New Zealand white rabbits were randomly divided into 4 groups: the normal group 6, the bone erosion Ning capsule group 18, the blank control group 18,Jian-Gu-Sheng group 18, and give the number. In the normal group of 6 rats were taken as the comparison standard, single cage rearing, the remaining 48 for hormone induced femoral head necrosis in rabbits model. After the modeling of Bone erosion ning capsule group(TG) according to the TG(normal 1±0.72mmol/L), bone erosion ning group takes 1.00mmol/L as a standard. The rabbits with a higher number than this are dialectically thought as blood stasis models, which will drench bone erosion ning No.1later. Rest of them is dialectically thought as kidney deficiency models, which will drench bone erosion ning No.2. positive control group with Jian-Gu-Sheng pills dissolved in distilled water regularly after intragastric administration, the blank control group using saline regularly gavage, respectively at fourth weeks, 8 weeks and 12 weeks were killed after removal bilateral femoral head embedded in paraffin and sectioned, using immunohistochemical method(SABC method) to detect the local expression of CD34、CYR61 and CTGF.2. To observe the effect of bone capsule on the morphology of the femoral head in rabbits with steroid induced avascular necrosis of the femoral head.After fourth weeks, 8 weeks and 12 weeks, the bilateral femoral head was removed and stained by HE, and the histological changes of the femoral head were observed under microscope.Experimental results:1.Expression determination of CD34、CYR61 and CTGFThe 4 weeks, the model group, positive control group and treatment group CD34、CYR61 and CTGF gray values were lower than those in normal group(P < 0.01), the treatment group and positive group of gray value is lower than that of model group(P <0.01); 8 weeks: treatment group and positive group of gray value is still lower than in the model group, the gray value, the treatment group of gray value is compared with the positive group of gray value and treatment group is low; the 12 week: normal group and positive group of gray values were higher than those in the treatment group(P < 0.01),the treatment group, the femoral head CD34、CYR61 and CTGF expression is higher than the expression of normal group and positive group(P < 0.01).2.Observation of HE dyeing4 weeks of normal femoral head trabecular structure normal, uniform morphology.Model group, treatment group and positive group of femoral trabecular bone compared with normal femoral head, the structure is irregular, uneven, and bone marrow fat cells than in normal femoral head increased.At 8 weeks showed the model group, the femoral head trabecular bone was thinning,the spacing increases, femoral head bone marrow fat cells increased significantly; bone lacunae number was increased significantly, while the treatment group and positive group shares bone trabecular structure has improved than that of the model group.At 12 weeks of femoral head slice showed normal bone trabecula structure integrity,model group micro structure without obvious changes, the treatment group and positive group, the bone trabecula structure becomes regular, more visible within the bone marrow hematopoietic cells, bone lacuna and the number of fat cells decreased compared with the previous, the total view treatment group, the femoral trabecular bone in the overall uniformity to be higher than that of the positive group.Experimental conclusion1. The combined use of Escherichia Coli Endotoxin and methylprednisolone can complete the model of rabbit steroid induced avascular necrosis of femoral head.2. Bone erosion Ning capsule can be enhanced CD34、CYR61 and CTGF expression,in favor of avascular necrosis of the femoral head in vascular remodeling and regeneration, bone necrosis of blood supply can be improved, necrotic bone repair in different degree, may be the drug of one of the important aspects of the mechanism of femoral head necrosis onset.3.Steroid induced necrosis of femoral vascular reconstruction and regeneration on the repair of femoral head has important significance.
