| Objective From the behavioral level,cell level and molecular level reveals that NTE promote the hippocampus nerve regeneration after cerebral ischemia reperfusion of endogenous neural cell apoptosis and its relationship with the Notch signal pathway..Methods 120 SD rats were randomly divided into three groups:sham operation group(A),model group(B) and drug group(C).The experiment using modified suture preparation of SD rat MCAO model,using the neurological function score,TTC staining,observe the establishment of rat model and pathological changes of ischemia reperfusion brain;Using immunohistochemical method to observe the sham group, model group, medication group 1 d,3 d,7 d and 14 d, four different time of the hippocampus DCX and compare the expression of change.Using Western Blot method to detect NICD expression changes in Notch signaling pathways at different time points.Result①The score of neurological evaluation:the scores in A were 0 from 1 d to 14 d. The scores in B and C were declined in 1 d to 14 d; compared with group A, in group C were not significant (P< 0.05) in 7 d.②TTC staining of brain tissue:there was no significant cerebral in group A; the infarct volume reduced significantly in group C from 3d to 14 d, which were significant (P <0.05) compared with group B at the same period. ③DCX and NICD:compared with the sham operation group, the expression of DCX and NICD were increased (P<0.05)in B and C; the expression of DCX and NICD increased significantly in group C, which were significant (P<0.05) compared with group B at the same period.Conclusion NTE can improve infarct volume and neurological deficits points and promote the regeneration of hippocampal neural stem cells in ischemic rat models of MCAO after reperfusion, and its mechanism is closely related to the Notch signal pathway. |
