Objective: To observe the effects of opposing needling on the proliferation of neural stem cells in rats with acute cerebral ischemia reperfusion injury, so as to explore the mechanism of the treatment of ischemic cerebrovascular disease.Methods: Fifty SD rats were randomly divided into three groups: blank group(10rats), sham operation group(10 rats), build module group(30 rats). The blank group was normal control group. The sham operation group was inserted 5mm thread to the internal carotid artery. The build module group used suture method prepared MCAO cerebral ischemia reperfusion injury model. After the rats wake up, base on the nerve function test scores, then randomly divided the build module group rats into model group, affected side acupuncture group and opposing needling group, 10 rats in each group.The blank group, sham operation group and model group were not treated,affected side acupuncture group and opposing needling group were treated by the corresponding acupuncture and electro-acupuncture treatment after modeling 24 h,treatment 7 days. At the end of the experiment, score the rats' neural function deficits scores, observe the rats' brain cortex structure by using light microscope, detect the Brd U positive cells and the average optical density value of Nestin in the ischemic side of hippocampus by immune tissue chemical and combined with image analysis, so as to evaluate the effect of acupuncture on neural stem cells after cerebral ischemia reperfusion injury.Result:1The neurological function scores : Build model groups rats were all appeared nerve function defect after modeling. After 7 days of treatment, compared with model group, the scores of nerve function defect decreased in both of opposingneedling group and affected side acupuncture group(P < 0.05, P < 0.01),and there was no difference in the scores between the opposing needling group and the affected side group(P < 0.01).2Brain tissue morphology and structure: Compared with the model group and affected side acupuncture group, the opposing needling group rat' brain cortex cell injury was lighter, the gap between the cell and the blood vessel decreased, and the capillary obviously increased.3 Proliferation of neural stem cells: Compared With the blank group and sham operation group, model group rat' Brd U positive cells and the average optical density value of Nestin increased(P < 0.01); compared with the model group, the Brd U positive cells and the average optical density value of Nestin in affected side acupuncture group and opposing needling group were increased(P < 0.01);opposing needling group over the affected side acupuncture group in Brd U and Nestin positive expression(P < 0.01).Conclusion: Opposing needling can improve the neural function after the rat'acute cerebral ischemia reperfusion injury, promote the proliferation of neural stem cells in the hippocampus. As the first step in the regeneration of nerve cells, the proliferation of neural stem cells can be differentiated into neurons, repair of nerve defect symptoms. The promotion of nerve stem cells proliferation may be one of the mechanisms of the effect of opposing needling in treatment of ICVD, but the inevitable connection need to further study.
