Objective: In recent years,the study found that Treg /Th17 and IL-17 are closely associated with the AR(Allergic Rhinitis).The thesis is to have a tentative research on the correlation of the expression of IL-17 and IL-35 in serum of AR in pre-specific immunotherapy,and to observe the changes of CD4+CD25+-regulatory T cells of sublingual immunotherapy.Methods: In this thesis,30 cases were randomly selected from outpatients of otolaryngological department in the second hospital of Hebei Medical university who were diagnosed of AR.After treatment,the same patients were as renamed as the post-therapy group.Another 30 cases were healthy subjects enrolled from physical examination branch of our hospital.All cases met grouping criteria.4.5ml peripheral blood was collected from the subjects,centrifuged as required.To collect the serum of upper layer,and detect the expression level of IL-35 and IL-17 in peripheral blood using double antibodies sandwich ELISA,the concentration of IL-35 and IL-17 according to OD value obtained.Contemporaneously,2.5ml peripheral blood of subjects was collected.The CD4+CD25+Foxp3+ T cell and CD4+IL-17+Th17 cell expression level were detected via flow cytometry,and the results were showed by the percentage of CD4+CD25+Foxp3+ T cells or CD4+IL-17+Th17 cell in CD4+ T cell.All experimental data were statistically analyzed by SPSS19.0 software.All data in this study belonged to the measurement data and testified as normal distribution via normality test data;AR group and the control group were compared with paired T-test.Correlation analysis was made using Pearson correlation test,with all statistics double side probability tested.Only when ?= 0.05,P<0.05,the test was considered statistically significant.Results:1 The expression of IL-17 in serum of AR before therapy was higher than the control group(21.5599.548 pg/mL),while the expression of IL-35 before therapy was less than the control group(29.4255.378 pg/mL).The percentage of CD4+CD25+Foxp3+ T cells in CD4+ T cell was lower than the one in control group(0.5430.178%).The percentage of CD4+IL-17+Th17 cells in CD4+ T cells was higher than the one in control group(0.3590.143%).The difference had a statistically significance.2 There was no obvious correlation between the expression of IL-17 and IL-35 in serum of AR before specific immunotherapy(r=-0.143,p>0.01);but there was a positive correlation between expression of IL-17 and the percentage of CD4+IL-17+Th17 cells in CD4+ T cells.3 Comparing the two groups,the expressions of IL-35 in pre-specific and post-specific immunotherapy group were respecitively 17.6727.458pg/mL and 25.133 9.122pg/mL,which indicated the difference has a statistically significance(t=-4.083,P<0.05).4 Comparing the two groups,the expression of IL-17 in pre-specific and post-specific immunotherapy group were respecitively 35.552=8.328pg/mL and 24.0708.409pg/mL which showed the difference has a statistically significance(t=5.030,P<0.05).5 The percentages of CD4+CD25+Foxp3+ T cells in CD4+ T cell from peripheral blood were 0.1470.069 in pre-specific immunotherapy group and 0.4040.200 in post-specific immunotherapy group(t=-10.584,P<0.05).6 The percentage of CD4+IL-17+Th17 cells in CD4+ T cells from peripheral blood were 1.1550.258% in pre-specific immunotherapy group and 0.7210.276% in post-specific immunotherapy group(t=6.258,P <0.05).7 The difference has a statistically significance between the control group and AR after therapy in the expression of IL-17,IL-35 and the percentage of CD4+CD25+Foxp3+ T cells,CD4+IL-17+Th17 cells in CD4+ T cells.Conclusions:Specific immunotherapy can have an impact on the expression levels of IL-17,IL-35 and also on Treg / Th17 cells balance in peripheral blood for patients with allergic rhinitis.
